Abstract: 

2Background: Carvacrol is a natural monoterpene phenolic compound that has biological activities with therapeutic applications, and this study aimed to investigate the apoptotic effect of Carvacrol on the human breast cancer cell line MDA-MB-231. Materials and Methods: After the preparation and cultivation of MDA-MB-231 cells, the IC50 value of Carvacrol on the cells was evaluated by the MTT assay method, and then the induction of apoptosis in the cell line treated with different concentrations of Carvacrol was observed by DAPI staining. The qPCR method was used to check the expression levels of Bax, P53, and Bcl-2 genes at the mRNA level. The results were analyzed using GraphPad Prism 9 software. Results: The results showed that the cytotoxicity of Carvacrol against MDA-MB-231 cancer cells was dose-dependent at 24 (P=0. 034) and 48 (P=0. 041) hours. The IC50 of Carvacrol at 48 h was 154. 2 μM. The MDA-MB-231 cells treated with Carvacrol showed induction of apoptosis from the mitochondrial pathway by increasing the expression of P53 and BAX genes and decreasing the expression of the anti-apoptotic Bcl-2 gene. In addition, the number of apoptotic cells with dense DNA increased significantly in the Carvacrol treatment group (P=0. 001). Conclusion: This study showed that treatment of the MDA-MB-231 cell line with Carvacrol can inhibit its growth and proliferation through the induction of apoptosis from the BAX/BCL-2 pathway. Considering the significant inhibitory effect of the treatment of cells with the Carvacrol compound, it seems that there is a suitable research field for using this compound in the control and treatment of breast cancer.

Abstract: 

Rheumatoid arthritis (RA) is one of the most common autoimmune diseases. Carvacrol, an important natural terpenoid product in aromatic plants such as thyme, has shown antiinflammatory effects in animal models of arthritis. However, its poor water solubility and high volatility have limited its application. In the present study in order to overcome this problem, we encapsulated carvacrol in the bovine serum albumin (BSA) nanoparticles and examined its therapeutic and immunomodulatory effects in adjuvant-induced arthritis (AIA). Carvacrol-loaded BSA nanoparticles were prepared by desolvation method. Nanoparticles had encapsulation efficiency (EE) of 67. 7 ± 6. 9% and loading capacity (LC) of 26. 6 ± 2%. The size of particles was 148 ± 25 nm and they had monomodal distribution. After arthritis induction, the rats were treated intraperitoneally with nanoparticle for every 3 days until day 28. The treatment of the rats with 375 mg/mL carvacrol-loaded BSA nanoparticle significantly decreased clinical severity score (27. 5 ± 9. 8%, p = 0. 008), erythrocyte sedimentation rate (33. 4 ± 10%, p = 0. 02), nitric oxide production (82. 3 ± 2. 6%, p = 0. 004) and interleukin (IL)-17 gene expression (55. 1 ± 8. 2%, p = 0. 003) compared to the untreated arthritic group. A higher reduction in inflammation severity in arthritic rats treated with carvacrol-loaded BSA in comparison to those treated with carvacrol alone was observed. In conclusion, encapsulation of carvacrol in nanoparticles reduced arthritis signs and release of NO and IL-17 inflammatory cytokine and therefore is suggested to be considered as a good approach for improving the therapeutic applications of carvacrol in RA.

Abstract: 

In our previous studies, the relaxant and anticholinergic, histamin (H1) inhibitory, and xantine-like activity effects of Carum copticum have been demonstrated on tracheal chains of guinea pigs. The relaxant effect of different fractions of essential oil from Carum copticum was also examined. Therefore the relaxant effect of two main costtuent substance of the plant (thymol and carvacrol) was examined in this study to find the possible responsible substance for the relaxant effact of this plant. The relaxant effects of three cumulative volumes (0.02, 0.04, and 0.08ml) of 1/100 diluted carvacrol were examined by their relaxant effects on pre-contracted tracheal chains of guinea pigs by 10 μM methacholine in two different conditions including: non-incubated tissues (group 1) and incubated tissues with 1 μM propranolol and 1 Μm chlorpheniramine (group 2), (for each group, n=5) and were compared with the effects of ethanol and theophylline. The results showed that relaxant effect of all three volumes (concentration) of carvacrol and were significantly higher than those of ethanol and even theophylline (p<0.05 to p<0.001). There was a positive correlation between relaxant effect of carvacrol and increment volumes. The relaxant effect of only first volume of carvacrol obtained in group 2 experiments was significantly greater than that of group 1 (p<0.05). These results indicated that the carvacrol has a potent relaxant effect on tracheal chains of guinea pigs which is not due to b2- adrenergic stimulatory or histamine H1 blocking effect.

Abstract: 

Background: Active packaging represents a defining strategy to improve food quality and safety of the packaged foods. This study aimed to evaluate the in vitro ability of commercial biopolymeric foams, namely Mater-Bi (MB), activated with 20% of carvacrol, to develop a completely biodegradable and compostable packaging to inhibit the growth of spoilage and pathogenic yeasts. Methods: MB foams, with and without carvacrol, were produced by melt mixing and the foaming process was performed in a laboratory press. The antifungal activity of foams containing carvacrol was tested applying the disk diffusion method. Statistical analysis was done using XLStat software version 7. 5. 2 for Excel. Results: Statistically significant differences (p<0. 05) were observed between sensitivity of the tested yeasts. Candida zeylanoides 4G362 and Rhodotorula mucilaginosa ICE29 were found to be the most sensitive strains with a clear zone of 28. 9± 0. 1 and 29. 0± 0. 1 mm, respectively. However, Aureobasidium pullulans was the least sensitive yeast strain, showing clear zone of 20. 4± 0. 3 mm. Conclusion: This study provided, for the first time, an in vitro analysis of the antifungal activity of MB foams activated with carvacrol against yeasts that commonly contaminate raw materials and processed foods. In conclusion, this biopolymer was highly effective against all the yeasts used as indicators strains.
