Abstract: 

Hepatocellular carcinoma is the fifth most common cancer worldwide, which is associated with severe complications. In this study, the effect of Avastin drug on the proliferation of HepG2 cells was investigated through the activation of the P53 pathway and the change of the antioxidant content of HepG2 cells cultured on a hydrogel scaffold containing HUVEC cells. In this study, HepG2 cells were cultured on type I collagen hydrogel scaffolds containing HUVEC and after treatment with different concentrations of Avastin, cell viability was measured using the MTT test and the rate of apoptosis using flow cytometry. The level of lipid peroxidation, superoxide dismutase (SOD) enzyme activity, total oxidant status (TOS) and total antioxidant capacity (TAS) were measured using calorimetric method and p53 protein level was measured using immunocytochemistry technique. Examining the results shows that the rate of apoptosis in HepG2 cells co-cultured with HUVEC-Scaffold and Avastin drug is significantly higher than other groups. The results show a significant increase in the amount of total oxidant and malondialdehyde and a significant decrease in the activity of the superoxide dismutase enzyme and total antioxidant capacity in the two groups of HepG2+Scaffold-HUVEC and HepG2+Scaffold treated with Avastin. Avastin can increase the percentage of p53 in HepG2 cells co-cultured with Scaffold and Scaffold-HUVEC. In this study, hydrogel scaffold was used as an option for optimal simulation of the life of cancer cells and it was shown that the drug Avastin has the ability to have a good anti-cancer effect in these structures.

Abstract: 

It was shown that several bioactive aromatic compounds with biological applications have the furan nucleus watch Numerous significant synthetic compounds include furan scaffold, which offers a helpful therapeutic idea and is found to have strong affinity for a range of receptors, assisting in the synthesis of novel, advantageous derivatives The antibacterial, antifungal, antiviral, anti-inflammatory, analgesic, antidepressant, antianxiolytic, anti-Parkinson, anti-glaucoma, muscle-relaxant, antihypertensive, diuretics, anti-ulcer, anti-aging, and anti-cancer effects of furan derivatives make them frequently utilized. Diverse furan derivatives have piqued the interest of researchers. Furan is a colourless liquid that boils almost at ambient temperature and is highly volatile and combustible. Electron-Rich Nature: Furan rings have the ability to engage in a variety of electrical interactions with biomolecules due to their electron-rich nature. This characteristic might make it easier to attach strongly to biological targets like enzymes or receptors, which would affect how they function. Aromaticity: Furan's aromatic properties may give the compounds stability, which could improve their metabolic stability and bioavailability. Numerous natural compounds and pharmaceutical molecules are known to depend heavily on aromatic systems for their bioactivity.Functional Group Diversity: A wide variety of derivatives can be synthesized by simply modifying furan scaffolds with different functional groups.

Abstract: 

Objective: The neurosphere assay is usually used to culture neural stem cells (NSCs). In this system, cells are under the least physical interaction with their environment. Using a more solid matrix might affect cellular behavior in terms of proliferation and differentiation that could benefit selection of cells with different properties. Alginate is a polysaccharide that is used in 3D cultures Here, we evaluated NSCs expansion in different alginate concentrations (0.25, 0.5, and 1%).Materials and Methods: First, NSCs from the E-14 mouse brain were cultured in different Alginate concentrations (0.25, 0.5 and 1%) and a control group. After 5-6 days, the cultures were evaluated for neurosphere growth and the sphere forming frequency was determined for each condition.Results: Evaluating the cultures from different groups showed that neurospheres could grow in all conditions but the sphere forming frequency was the highest in control comparing to the Alginate groups. In Alginate groups, it was revealed the higher the Alginate concentration, the lower the sphere forming frequency. Furthermore, all spheres from both control and Alginate groups could be serially passaged. Comparing the size of spheres, it was evident that the spheres in higher concentration of Alginate were larger than the control.Conclusion: Our study shows that the mechanical effects of alginate 3D scaffold reduce the sphere forming frequency of neural stem cells. At the same time growing NSCs in Alginate scaffold results in larger spheres that could be due to selection of bona fide NSCs in Alginate culture comparing to the control.

Abstract: 

Background: The goal of tissue engineering is to create biological solutions that restore, maintain, and improve the function of damaged tissue. Scaffolds are structures based on extracellular matrix materials that have undergone various treatments. Objectives: This study aimed to prepare polycaprolactone/silymarin and polycaprolactane/tragacanth scaffolds and compare the morphology and viability of PC12 cell lines. Methods: A 7% polycaprolactane solution (dissolved in acetic acid) was mixed with a 0.9% silymarin solution to prepare the polycaprolactane scaffold and silymarin loading, and a 7% polycaprolactane solution was mixed to prepare the polycaprolactane and tragacanth scaffold. Tragacanth solution was mixed at a concentration of 0.7% by weight, and then two scaffolds were prepared by electrospinning. The morphology of the scaffolds was studied by scanning electron microscopy (SEM), and the chemical structure of the scaffolds was studied by ATR-FTIR spectroscopy. The biocompatibility of the scaffolds and cell survival of PC12 cells was investigated by MTT assay. Results: The morphology of the scaffolds and their chemical structure showed good porosity and successful loading of silymarin onto PCL and a suitable combination of tragacanth with PCL. The biocompatibility of the scaffolds was evaluated at 24, 48, and 72 hours after PC12 cell culture. Cell survival was found to increase on polycaprolactane/silymarin scaffolds compared to polycaprolactane/tragacanth. Conclusions: From the results of this study, loading polycaprolactane scaffold with silymarin increases the proliferation and survivability of PC12 cells compared to polycaprolactane/supporting scaffold, which may be a good candidate for neural tissue engineering.

Abstract: 

TREATMENT OF FULL-THICKNESS SKIN WOUNDS WITH MINIMAL SCARRING AND COMPLETE RESTORATION OF NATIVE TISSUE PROPERTIES STILL EXISTS AS A CLINICAL CHALLENGE. WE FABRICATED A BILAYER SKIN SUBSTITUTE BY COATING HUMAN AMNIOTIC MEMBRANE WITH AN ELECTROSPUN NANOFIBROUS SILK FIBROIN LAYER TO IMPROVE MECHANICAL PROPERTIES OF THE AMNIOTIC MEMBRANE (AM). SINCE THIS BILAYER SCAFFOLD EXHIBITED IMPROVED MECHANICAL PROPERTIES, SURFACE HYDROPHILICITY, AND IN VITRO BIOCOMPATIBILITY, IN VIVO BIOLOGICAL BEHAVIOR OF THIS SCAFFOLD WAS INVESTIGATED IN MURINE FULL-THICKNESS SKIN WOUND MODEL. DONUT-SHAPED SILICON SPLINTS WERE UTILIZED TO PREVENT WOUND CONTRACTION IN MOUSE SKIN AND SIMULATE RE-EPITHELIALIZATION, WHICH IS THE NORMAL PATH OF HUMAN WOUND HEALING. THE SKIN REGENERATION USING BILAYER SCAFFOLD WAS COMPARED WITH AM AND UNTREATED DEFECT AFTER 30 DAYS. TISSUE SAMPLES WERE TAKEN FROM HEALED WOUND AREAS AND STUDIED BY MEANS OF HISTOPATHOLOGICAL EVALUATION AND IMMUNOHISTOCHEMICAL STAINING, TO VISUALIZE INVOLUCRIN (IVL), P63, COLLAGEN I, CD31, AND VEGF. IN ADDITION, MRNA EXPRESSION OF IVL, P63, INTERLEUKIN-6 (IL-6) AND CYCLOOXYGENASE-2 (COX-2) WAS STUDIED. BASED ON THE OBTAINED DATA, THE APPLICATION OF BILAYER SCAFFOLD RESULTED IN THE BEST EPIDERMAL AND DERMAL REGENERATION, DEMONSTRATED BY HISTOPATHOLOGICAL EXAMINATION AND MOLECULAR ANALYSES. THE MRNA EXPRESSION LEVELS OF INFLAMMATORY MARKERS (IL-6 AND COX-2) IN REGENERATIVE TISSUES BY BILAYER SCAFFOLD WERE DOWN-REGULATED AND EXPRESSION PATTERN OF KERATINOCYTE MARKERS INCLUDING IVL AND P63 MARKERS AT BOTH MRNA AND PROTEIN LEVELS IN BILAYER SCAFFOLD GROUP WAS MORE SIMILAR TO NATIVE TISSUE IN COMPARISON TO AM AND NO-TREATMENT GROUPS. THERE WAS NO SIGNIFICANT DIFFERENCE IN EXPRESSION LEVEL OF COLLAGEN I, CD31, AND VEGF AMONG DIFFERENT GROUPS. BASED ON ANIMAL STUDY RESULTS, PRELIMINARY EVALUATION OF WOUND HEALING USING THIS CONSTRUCT WAS DONE IN 10 PATIENTS WITH FOOT ULCER AFTER SIGNING INFORMED CONSENT. THE DATA SHOWED SAFETY AND EFFICACY OF THE SCAFFOLD AS WELL AS WOUND AREA WAS SIGNIFICANTLY DECREASED FROM THE BEGINNING OF TREATMENT TO THE END OF THE FIRST FOUR WEEKS. STUDIES DONE IN DIFFERENT PHASES WERE APPROVED BY MEDICAL ETHIC COMMITTEE OF AVICENNA RESEARCH INSTITUTE (ACECR, TEHRAN, IRAN) AND REGISTRATED IN IRANIAN REGISTRY OF CLINICAL TRIALS. CONCLUSIVELY, THESE PROMISING RESULTS SUGGEST THAT THIS BILAYER SCAFFOLD CAN REPRESENT A POTENTIAL SUBSTITUTE FOR SKIN REGENERATION APPLICATION AND SERVE AS SUPPORTING EVIDENCE FOR PROCEEDING TO MAJOR CLINICAL PHASE.

Abstract: 

Background. Due to some limitation and problems of implant and GBR, tissue engineering has become more commonly used. 3D-printed scaffolds are investigated in periodontal applications shows outcomes of success. (3D) scaffold architecture closely mimics native (ECM). It enhances cell adhesion, proliferation, differentiation and tissue regeneration. Methods. 3D scaffold fabrication: CAD/CAM, CAD models are images from computed tomography (CT) With the development of SFF. Multiphasic scaffold contains channel-like “ fiber-guiding architecture” of the PDL, which are functionally oriented and integrated. Compartmentalization can prevent tooth ankylosis. there are two Methods. (1) cell seeding into a “ prefabricated” scaffold, and (2) cell encapsulation during scaffold fabrication, which protectscells from immune system. Results. PLC is moldable for required shapes and have good mechanical properties but it has two undesirable points: slow degradation rate and release acidic byproduct for solving these two problems and prevention of scaffold exposure, PLC is combined with bioceramics, which neutralize the acidic byproducts of PLC, and increase rate of degradation. Conclusions. scaffold facilitate cell penetration, vascularization. pore network prevents core necrosis. In alveolar bone regeneration, augmentation, and socket preservation, scaffolds made of bioceramics can be recommended. But because of their weak mechanical properties, bioceramics should combined with strong biomaterials as mentioned earlier and PLC should use with bioceramics. In non-load-bearing areas, collagen could be preferred biomaterial. bioceramic/collagen mix is closest replicate of the ECM composition of native bone. combination of collagen with hydroxya-patiteis encouraged in bone tissue regeneration Much work needs for promising results, clinically.
