Abstract: 

Effect of bupropion, an antidepressant drug, on sniffing behavior has been examined in male rats. Animals treated intraperitoneally (i.p.) with different doses of bupropion (5-40 mg/kg), showed sniffing behavior in a dose-related manner. Pretreatment of animals with the dopamine antagonists, SCH 23390 (0.025-0.1 mg/kg, Lp.) or sulpiride (12.5-50 mg/kg, i.p.) decreased the bupropion induced sniffing. Reserpine pretreatment (2.5 mg/kg, i.p,18h) alone and in combination with alpha-methyl-p-tyrosine (AMPT; 250 mg/kg, i.p.) also reduced the bupropion induced sniffing. The α-adrenoceptor antagonist, phenoxybenzamine (5 and 10 mg/kg, i.p.), the padrenoceptor antagonist, propranolol (5 and 10 mg/kg, Lp.) did not affect the bupropion induced response. The antimuscarinic, atropine (5 and 10 mg/kg, i.p.), increased the response significantly. It is concluded that bupropion induces sniffing through indirect dopaminergic mechanism.

Abstract: 

We present a case report of bupropion-induced diplopia in a 26-year-old woman who suffered from atypical depression. After four weeks of taking bupropion (225 mg per day) she complained of headaches, blurred vision, and diplopia. No neurological and ophthalmologic abnormal signs were found. Her brain magnetic resonance imaging (MRI) and magnetic resonance venography (MRV) were normal as well. After tapering off and discontinuing bupropion, her diplopia resolved within a week.

Abstract: 

This study was designed determine the efficacy of bupropion versus placebo in subjects with negative symptoms of schizophrenia.A convenience sample of 40 patients of both genders aged 18-60 years who were living in psychiatric care centers were randomly treated with bupropion (started with 75 mg twice a day; increased to 100 mg thrice daily) or placebo. The diagnosis of schizophrenia was confirmed by a psychiatrist based on Diagnostic and Statistical Manual of Mental Disorders, Fourth Edition, Text Revision (DSM-IV-TR) criteria. Before and after the intervention, severity of negative symptoms was determined using a reliable and valid Persian version of Scales for the Assessment of Negative Symptoms (SANS).Comparison of post-treatment total SANS score and subscale scores between bupropion treated patients and placebo group demonstrated no significant difference. Moreover, comparison of pre- treatment and post-treatment total SANS score and subscales within 2 groups revealed that nor bupropion neither placebo improved the severity of negative symptoms significantly.Present study demonstrated that bupropion has no significant effect on SANS score of patients with severe negative symptoms. However, further studies with larger sample size are recommended to achieve more accurate results.

Abstract: 

Antidepressants are widely used for the treatment of various neuropathic pain conditions in humans. Recent studies have demonstrated that bupropion is effective for the treatment of neuropathic pain. Also antidepressants like bupropion showed anti-inflammatory properties. So in the present study, the analgesic and anti-inflammatory effects of bupropion in mice and rat were investigated. The acetic acid, formalin and hot plate tests were used in male mice to assess analgesic activity. For evaluation of anti-inflammatory effect, carrageenan-induced rat paw edema and croton oil-induced ear edema were used. Bupropion was administered at the doses of 10, 20 and 40 mg/kg (i.p.). Bupropion at a dose of 40 mg/kg significantly reduced acetic acid-induced abdominal writhes and also was effective in suppression of formalin-induced behavior and showed significant analgesia in hot plate test. While 40 mg/kg bupropion showed considerable anti-inflammatory response in carrageenan test, but no effect was observed in croton oil-induced ear edema. The results showed that bupropion has analgesic and anti-inflammatory effects in animal models and further studies are needed to find out its mechanism of action.

Abstract: 

Bupropion, as dopamine reuptake blocker and an antidepressant agent, has been previously shown to attenuate memory performance in some doses. The aim of this study was to investigate active avoidance learning in rat. This study was designed to test the effect of intra-hippocampal bupropion injection on active avoidance learning.Adult Wistar rats (200-250gr) were tested for learning behavior in two way active avoidance in the shuttle-box. The conditioned stimulus was light that located on the ceiling of dark part and unconditioned stimulus was an electric foot shock (max 1-2mA, 10 sec) delivered through stainless steel rods covering the floor.The conditioned-unconditioned stimulus interval was 5 sec. One trial was limited to 20 sec, and was repeated on 5 consecutive days. Sessions were always started at the same time of day. If following conditioned stimulus the rat changed the compartment within the first 5 sec, and then a conditioned reaction and avoidance, respectively, was completed. The number of conditioned reactions (reaction time, 5 s) was recorded for evaluation of the learning performance. Drug injected by stereotaxic implanted guide cannulae. The rats were trained (20 trails, 2 times in each day) of a two way active avoidance task. After completion learning process (5 days), rats received bupropion (5 mg/μl/1- 2 min) 30 min before testing. The learned process was checked again with same paradigm.The results were analyzed as before and after pulled parameters. The results showed that intra-hippocampal bupropion can deficit memory performance. Bupropion impaired memory performance at least 20% in treated period. It decreased the number of avoidance reactions and increased reaction time also. Although bupropion is a dopamine reuptake blocker but it concludes that bupropion can bind to other receptors such as Acetylcholine receptors and interfere learning and memory.
