Abstract: 

Background: Mycobacterium bovine, as the etiology of bovine TB, involves human in some cases and with Mycobacterium tuberculosis are regarded as global health problems. The aim of this study was to extract and compare protein profile of these strains in order to achieve effective biomarkers for diagnosis and the vaccine components.Materials and methods: First, the clinical samples were cultured on Lowenstein-Jensen (LJ) medium by the N-acetyl-L-cysteine-sodium hydroxide method and the biochemical tests and antibiotic susceptibility were used.Colonies were grown in the Middlebrook 7H9 medium and, upon harvesting the new colonies, secretory and membrane proteins were extracted by sonication and ammonium sulfate and alcohol precipitation. Concentration was determined by Bradford method and finally the comparison was made through one dimensional electrophoresis.Results: The major discrepancy between two strains of Mycobacterium tuberculosis and Mycobacterium bovis was 45 and 60 KDa bands as well as the zone between 14 and 45 KDa bands of secretory proteins, in the banding separation membrane proteins.Conclusion: Discrepancy in the protein bands of sensitive strains and Mycobacterium bovis seems to be used as an effective protein marker or even a biomarker in distinguishing the sensitive and bovis strains; also similarities can be used for immunization purposes.

Abstract: 

Background: Recently it is shown that Mycobacterium tuberculosis Beijing genotype are associated with drug resistance. Thus a simple and rapid method is required for identification and differentiation of Beijing family isolates. Therefore the aim of this study is to evaluate discriminatory power of different loci in both 12 MIRU-VNTR and 15 MIRU-VNTR Methods for Beijing genotype.Methods: In total 105 Mycobacterium tuberculosis strains were identified by spoligotyping, then genomic analysis was carried out by using 12 and 15 MIRU-VNTR methods. Allelic diversity for each locus was calculated using Hunter Gaston Discriminatory Index (HGI).Results: With spoligotyping 20 out of 105 isolates (19.05%) belonged to the Beijing family. Investigating all strains with 12 and 15 MIRU-VNTR showed that 11 loci were highly discriminative (HGI ³0.6) and QUB26 had the highest discriminatory power (HGI=0.84). Number of repeats in QUB11b was specific for Beijing genotype, and only MIRU16 was highly discriminator for this genotype. QUB26, Mtub21, MIRU39, MIRU27, MIRU23 and MIRU26 were moderately discriminator (0.4£HGI<0.6) and other loci were poorly discriminator for Beijing strains (HGI<0.4).Conclusion: 12 and 15 locus MIRU-VNTR are simple and rapid methods and suitable for differentiation of Beijing genotype from other MTB strains, however these two methods are not suitable for discriminating Beijing family among themselves. In overall discriminatory power of 15 locus MIRU-VNTR method was higher than 12 locus MIRU-VNTR.

Abstract: 

Background: tuberculosis (TB) is an infectious disease caused by Mycobacterium tuberculosis.Objectives: Development of the drug resistance is becoming a threat to disease control, which underscores need for new agents targetingM. tuberculosis.Materials and Methods: In this study, analysis of gene expression was performed using real-time polymerase chain reaction (RTPCR).Results: Results of the current study showed that the minimum inhibitory concentration value of cinnamaldehyde against M. tuberculosis was 200mg/mL. Moreover, RT-PCR data showed that a total of 25 genes were regulated by the cinnamaldehyde. Of these, 12 genes were up-regulated, and 13 genes were down-regulated.Conclusions: Cinnamaldehyde is a pattern to expand the new anti-TB drugs, because the targets of the cinnamaldehyde are different from those of anti-tubercular agents.
