Abstract

 BACKGROUND AND AIM: STUDIES HAVE PROVED THAT SUSPENSION CONTAINING NANOPARTICLE FORMULATIONS OF ANTIMICROBIAL AGENTS BASED ON SOME FUNDAMENTAL FEATURES OF NANOPARTICLES HAVE BETTER ANTIMICROBIAL EFFECT. VANCOMYCIN IS A GLYCOPEPTIDE ANTIBIOTIC WHICH POSES ACTIVITY AGAINST GRAM POSITIVE BACTERIA. ENTEROCOCCUS IS A GENUS OF BACTERIA THAT BECAME RESISTANT TO A WIDE RANGE OF ANTIBIOTICS IN LAST DECADES, AND CAUSE SEVERE INFECTIONS IN HOSPITALIZED PATIENTS. THE PURPOSE OF THIS RESEARCH WAS TO PREPARE VACOMYCIN NANOPARTICLES USING POLY (LACTIC-CO-GLYCOLIC ACID) POLYMER AND TO COMPARE THE EFFECTIVENESS OF NANOPARTICLES IN COMPARISON WITH DRUG SOLUTION AGAINST CLINICAL ENTEROCOCCUS ISOLATES.METHODS: VANCOMYCIN-LOADED PLGA NANOPARTICLES WERE FREEZE DRIED AFTER PREPARATION BY W1/O/W2 SOLVENT EVAPORATION METHOD USING DIFFERENT RATIOS OF DRUG TO POLYMER (1: 0.5, 1: 1 AND 1: 2). ENTEROCOCCUS SPECIES ISOLATED FROM CLINICAL SAMPLES (IN TWO, SUSCEPTIBLE AND RESISTANT TO VANCOMYCIN, GROUPS). THE MIC VALUES OF VANCOMYCIN-LOADED NANOPAETICLES AND VANCOMYCIN SOLUTION WERE DETERMINED USING BROTH MACRO DILUTION METHOD. THE OBTAINED MIC VALUES FOR DRUGLOADED NANOPARTICLES COMPARED TO THOSE OF DRUG SOLUTION….
