Abstract

 BACKGROUND AND AIM: ANTIBIOTIC RESISTANCE RATES ESPECIALLY AGAINST FLUOROQUINOLONES IN UROPATHOGENIC E.COLI ARE RAPIDLY RISING. THE AIM OF THIS STUDY WAS TO INVESTIGATE THE FREQUENCY OF QNRA AMONG E. COLI STRAINS ISOLATED FROM URINARY INFECTIONS.METHODS: WE EXAMINED ALL E. COLI STRAINS ISOLATED FROM URINARY INFECTIONS IN EMERGENCY UNIT IN BAQIYATALLAH HOSPITAL, TEHRAN. BACTERIAL STRAINS WERE ISOLATED, DETECTED AND IDENTIFIED BY STANDARD MICROBIOLOGICAL TESTS. TO SCREEN THE FLUOROQUINOLONE-RESISTANT ISOLATES, THE ANTIMICROBIAL SUSCEPTIBILITY TESTING WAS DETERMINED ACCORDING TO KIRBY BAUR ASSAY. DNA WAS EXTRACTED BY BOILING METHOD. ALL STRAINS WERE EXAMINED FOR THE PRESENCE OF QNRA GENE BY PCR. THE PCR AMPLICONS WERE VISUALIZED AFTER ELECTROPHORESIS AND STAINED WITH ETHIDIUM BROMIDE.RESULTS: ONE HUNDRED E. COLI STRAINS WERE ISOLATED AND INCLUDED IN THE STUDY. ANTIBIOTIC SUSCEPTIBILITY TESTING SHOWED THAT 35.45% AND 65% WERE SUSCEPTIBLE TO NALIDIXIC ACID AND CIPROFLOXACIN, RESPECTIVELY.1% OF THE ISOLATES HARBORED QNRA GENE…..
