Abstract

 COCKLEBUR (XANTHIUMSTRUMARIUM L.) IS AN ANNUAL WEED THAT CAN CAUSE SEVERE LOSSES INMANY CROPS WORLDWIDE. PLANT PATHOGENS HAVE BEEN USED AS BIOHERBICIDES FOR THE CONTROL OF THE WEED. A SEVERE BACTERIAL DISEASE WITH LEAF SPOTS SURROUNDED WITH YELLOW HALO SYMPTOM HAS BEEN OBSERVED ON COCKLEBUR IN NORTHWESTERN AREAS OF IRAN. THE OBJECTIVE OF THIS STUDY WAS TO IDENTIFY AND CHARACTERIZE THE PATHOGEN BY PHENOTYPIC, GENETIC AND PATHOGENICITY TESTS. SAMPLES WERE COLLECTED FROM DISEASED PLANTS IN VARIOUS NORTHWESTER AREAS DURING 2011-2012. IN THE LAB, ISOLATION OF BACTERIA FROM THE SAMPLES WAS PERFORMED ON CULTURE MEDIA. PALE YELLOW COLONIES CONSISTENTLY WERE ISOLATED FROM THE SAMPLES. TWENTY ISOLATES WERE SELECTED FROM CULTURE MEDIA AND WERE SUBJECTED TO PHYSIOLOGICAL AND BIOCHEMICAL TESTS, REP-PCR, 16S RRNA SEQUENCING AND PATHOGENICITY TESTS. BIOCHEMICAL TESTS IDENTIFIED COCKLEBUR STRAINS AS XANTHOMONAS SP.. REP-PCR FINGERPRINTING PLACED ALL THE PATHOGEN STRAINS IN ONE CLUSTER WITH LOW GENETIC VARIABILITY WITHIN THE STRAINS. BASED ON COMPARISON OF THE 16S RRNA SEQUENCES WITH THE NATIONAL CENTRE FOR BIOTECHNOLOGY INFORMATION (NCBI) TAXONOMY DATABASE, COCKLEBUR STRAINS BELONG TO THE GENUS XANTHOMONAS. IN PATHOGENICITY TESTS, ALL COCKLEBUR AND PEA PLANTS INOCULATED BY THE PATHOGEN STRAINS DEVELOPED SYMPTOMS OF THE DISEASE AND FROM ALL DISEASED SAMPLES; BACTERIAL STRAINS WERE RECOVERED ON NA MEDIUM. MORE STUDY IS NEEDED TO DETERMINE EXACT SYSTEMATIC SITUATION OF THE PATHOGEN AMONG KNOWN XANTHOMONADS, AND ALSO TO EXAMINE VIRULENCE AND HOST RAGE OF THE PATHOGEN. THIS STUDY WILL CONTRIBUTE TO DEVELOP A NEW BIO_HERBICIDE TO CONTROL COCKLEBUR WEED.
