Abstract

SEPTORIA LEAF BLOTCH (STB) IS ONE OF THE MOST DESTRUCTIVE DISEASES OF WHEAT IN ALL WHEAT GROWING AREAS WORLDWIDE. MYCOSPHAERELLA GRAMINICOLA CAUSES THIS DISEASE IN WHEAT. THE USE OF RESISTANT CULTIVARS IS THE MOST EFFECTIVE, ECONOMICAL AND ENVIRONMENTALLY SAFE METHOD OF MANAGING STB. FOR SUCCESSFUL BREEDING OF RESISTANT CULTIVARS, OUR KNOWLEDGE ABOUT WHEAT - M. GRAMINICOLA INTERACTION AND GENETIC RESISTANCE IS SIGNIFICANTLY IMPORTANT. IN THIS STUDY, CDNA-AFLP ANALYSIS WAS USED TO IDENTIFY GENES INVOLVED IN RESISTANCE TO STB. RESISTANT CULTIVAR WANG SHUI BAI AND SUSCEPTIBLE CULTIVAR FALAT WERE INCUBATED WITH M. GRAMINICOLA IN THE GREENHOUSE, AND THEN WERE SAMPLED AT 11 TIME POINTS. USING 37 PRIMER COMBINATIONS, 2200 FRAGMENTS WERE OBSERVED. 59 TRANSCRIPT-DERIVED FRAGMENTS THAT SHOWED DIFFERENTIAL EXPRESSION AFTER INOCULATION WERE IDENTIFIED, SEQUENCED AND CHARACTERIZED. FINALLY, 32 ESTS RELATED TO STB WERE SUBMITTED AT NCBI DATABANK. THESE RESULTS DETERMINED SOME GENES INVOLVED IN STB RESISTANCE IN WHEAT DURING INFECTION WHICH CAN HELP TO ELUCIDATE GENETICS BASIS OF RESISTANCE.
