Abstract

 BACKGROUND AND AIM: PROTEIN AGGREGATION WAS FIRST DISCOVERED IN MAMMALIAN CELLS SUCH AS ALZHEIMER DISEASE. THIS TOPIC PROPOSED AN INTERESTING IDEA THAT THE SAME BASIC MECHANISM THAT CAN CAUSE NEURONAL DEATH CAN BE USED AS A STRATEGY TO CONFLICT BACTERIAL INFECTION.METHODS: 125 PEPTIDES BASED ON THE PROTEOME OF ESCHERICHIA COLI O157: H7 WERE DESIGNED. THE MICS OF AGGREGATOR PEPTIDES WERE DETERMINED BY BROTH MICRODILUTION ACCORDING TO EUCAST. THE KILLING KINETIC WAS MEASURED BY CHANGES IN THE VIABLE COUNTS OF BACTERIA. SEM AND TEM WERE USED TO INVESTIGATE MORPHOLOGICAL CHANGES. IN ORDER TO EVALUATE THE TARGET SPECIFICITY FOR THE ACTIVE PEPTIDES, HETEROLOGOUS AND OVER-EXPRESSION OF THE TARGETED PROTEIN WERE PERFORMED TO OTHER STRAINS. TOXICITY TEST IN VITRO AND IN VIVO WERE ASSESSED. THE IN VIVO ACTIVITY OF THE AGGREGATORS ON MICE WAS TESTED...
