Abstract

 INTRODUCTION: TISSUE ENGINEERING HAS BEEN DEVELOPED USING NON-LIVING CONSTRUCTS SUCH AS CELL-FREE SPONGES, SHEETS AND FIBROUS MATS. WITH THE INTRODUCTION OF CELLS, SPECIALLY STEM CELLS, TISSUE-ENGINEERED PRODUCTS BECAME CAPABLE OF PROVIDING HEALTHY CELLS TO THE INJURED SITE. FOR THIS AIM, THERE ARE TWO CELL DELIVERY APPROACHES LIKE SCAFFOLD-BASE AND SCAFFOLD-FREE. IN CONTRAST TO THE FIRST-GENERATION TISSUE ENGINEERING, SCAFFOLD-BASE METHOD, THAT USES BIODEGRADABLE POLYMERS MIMICKING THE EXTRA CELLULAR MATRIX FOR LIVING CELLS, A NOVEL INTELLIGENT TEMPERATURE-RESPONSIVE SCAFFOLD-FREE CULTURE SURFACE WAS DEVELOPED AS A NEXT GENERATION TISSUE ENGINEERED METHODOLOGY AND CALLED ‘CELL SHEET ENGINEERING’. SINCE CELL SHEETS CAN BE HARVESTED WITHOUT PROTEASE TREATMENT, THE CELL–CELL AND CELL-ECM JUNCTIONS, WHICH ARE SUSCEPTIBLE TO PROTEASE TREATMENT, ARE PRESERVED WITHOUT DAMAGE. MOREOVER, 3-D TISSUE CAN BE MADE BY LAYERING THE CELL SHEETS WITHOUT THE USE OF SCAFFOLDS, TO FORM A BIOLOGICALLY FUNCTIONAL TISSUE...
