Abstract

 OBJECTIVE: IN THIS STUDY, WE HAVE INVESTIGATED THE STABILITY OF C-MYC AT VARIOUS PH AND IONIC STRENGTHS.MATERIALS AND METHODS: BY FLUORESCENCE SPECTROSCOPY, UV-VISIBLEAND CIRCULAR DICHROISM (CD) TECHNIQUES. PROTEINS OF STEM CELLS ARE IMPORTANT IN HUMAN’S MATURITY. WE STUDIED ONE OF THE TRANSCRIPTION FACTORS OF STEM CELLS (C-MYC) WHICH IS A CELLULAR ONCOPROTEINAND GENERALLY UP REGULATED IN MOST OF HUMAN CANCERS. ALTERATIONS OF CHROMOSOME 8, PREFERENTIALLY DELETIONS OF 8P AND GAINS OF 8Q, BELONG TO THE MOST FREQUENT CYTOGENETIC CHANGES IN BLADDER CANCERS. PROTEIN C-MYC ON 8Q24 IS A CANDIDATE ONCOGENE IN THIS REGION. THIS FACTOR HAS A CRITICAL ROLE IN THE STEM CELLS AND ECTOPIC EXPRESSION OF C-MYC AND OCT4 CAUSE A PLURIPOTENT STATE UPON SEVERAL DIFFERENTIATED CELL TYPES AND GENERATING INDUCED PLURIPOTENT STEM CELLS (IPSCS). THE FLUORESCENCE OF C-MYC IS CHARACTERIZED AT LEX=280 NM, 295 NM AND 305 NM. BY PH ALTERNATIVE, FLUORESCENCE INTENSITY INCREASE CAN BE MEASURE STABILITY PARAMETERS OF C-MYC...
