Abstract

 INTRODUCTION: TO COMPARE NORMAL AND LEUKEMIC PROCESSES, AN UNDERSTANDING OF NORMAL STEM CELL MAINTENANCE IN THE BONE MARROW IS NECESSARY. HEMATOPOIETIC STEM CELLS (HSCS) PRINCIPALLY EXIST WITHIN A TIGHTLY REGULATED ENVIRONMENT IN THE BONE MARROW, AND THIS ENVIRONMENT IS COMMONLY REFERRED TO AS THE NICHE. WITHIN THE NICHE, MOST HSCS ARE QUIESCENT, AND ENTRY INTO THE CELL CYCLE IS REGULATED BY CONTACT-DEPENDENT AND INDEPENDENT MECHANISMS. THE STEM CELL NICHE HAS BEEN EXPLOITED IN THE SETTING OF HSC TRANSPLANTATION AND HIJACKED BY MALIGNANCIES, SUCH AS LEUKEMIA. HOMING AND ENGRAFTMENT TO THE NICHE MAY BE ADDITIONAL TARGETS IN THE THERAPY OF LEUKEMIA...
