EFFECTS OF THYROID STATUS ON INSULIN SECRETION IN RAT PANCREATIC ISLETS AND ROLE OF KATP CHANNELS

Abstract

 INTRODUCTION: SINCE THYROID HORMONES PLAY IMPORTANT ROLE IN CARBOHYDRATE METABOLISM, DISTURBANCES OF THYROID FUNCTION CAN AFFECT GLUCOSE HOMEOSTASIS. THIS STUDY EXAMINED THE EFFECTS OF THYROID HORMONE DEFICIENCY AND EXCESS ON GLUCOSE TOLERANCE, INSULIN SECRETION AND KATP CHANNEL SENSITIVITY IN ISOLATED PANCREATIC ISLETS.METHODS: THE MALE WISTAR RATS WERE DIVIDED INTO THREE GROUPS: CONTROL, HYPERTHYROID AND HYPOTHYROID RATS. THE CONTROL GROUP CONSUMED WATER, WHILE THE HYPOTHYROID AND HYPERTHYROID GROUPS RECEIVED RESPECTIVELY WATER CONTAINING 0.03% METHIMAZOLE AND 10 MG PER LITER LEVOTHYROXINE. THESE TREATMENTS WERE ADMINISTERED FOR 4 WEEKS. AN INTRAPERITONEAL GLUCOSE TOLERANCE TEST (IPGTT) WAS CARRIED OUT ON EXPERIMENTAL ANIMALS. IN IN VITRO STUDIES, INSULIN CONTENT, GLUCOSE INDUCED INSULIN SECRETION AND THE SENSITIVITY OF KATP CHANNELS TO GLIBENCLAMIDE AND DIZOXIDE WERE COMPARED.RESULTS: IN COMPARED WITH THE CONTROLS, IN BOTH HYPOTHYROID AND HYPERTHYROID RATS, PLASMA GLUCOSE LEVEL DURING IPGTT WAS INCREASED AND PLASMA INSULIN LEVEL AND GLUCOSE INDUCED INSULIN SECRETION IN ISOLATED PANCREATIC ISLETS WERE DECREASED. IN HYPERTHYROID, BUT NOT IN HYPOTHYROID RATS INSULIN CONTENT OF THE ISOLATED ISLETS WERE SIGNIFICANTLY LOWER THAN THAT IN CONTROL GROUP. RESPONSE OF HYPOTHYROID ISLETS TO GLIBENCLAMIDE AND DIAZOXIDE WAS SIGNIFICANTLY LOWER THAN THAT IN THE CONTROL AND HYPERTHYROID ISLETS.CONCLUSION: RESULTS SHOWED THAT THYROID DYSFUNCTION LEADS TO GLUCOSE INTOLERANCE AND IMPAIRED INSULIN SECRETION. THESE FINDINGS ALSO SHOWED THAT REDUCED RESPONSE OF ISLETS TO GLIBENCLAMIDE AND DIZOXIDE IN HYPOTHYROID RATS CAN RESULT FROM REDUCTION IN SENSITIVITY OR ACTIVITY OF KATP CHANNELS.
