PERK AND C-FOS EXPRESSION PATTERN IN THE LUMBAR SPINAL CORD NEURONS OF CONGENITAL HYPOTHYROID WEANED PUPS FOLLOWING FORMALIN-INDUCED PAIN

Abstract

 INTRODUCTION: THYROID HORMONE IS INVOLVED IN THE NEUROPLASTIC MECHANISMS OF NOCICEPTIVE CIRCUITS. IN ADDITION, HYPOTHYROID RATS ARE HYPOSENSITIVE TO TONIC NOCICEPTION (FORMALIN PAIN) WITH THE ABSENCE OF LICKING BEHAVIOR (PHASE II).THE EXPRESSION PATTERN OF DORSAL HORN EXTRACELLULAR SIGNAL-REGULATED KINASES (ERK1/2) AND IMMEDIATE EARLY GENES (C-FOS) IN HYPOTHYROID WEANED PUPS DURING FORMALIN PAIN PROCESSING IS THE AIM OF PRESENT STUDY.METHODS: PREGNANT DAMS WERE GIVEN 0.005 G / 100ML OF ANTI-THYROID DRUG PROPYLTHIOURACIL IN DRINKING WATER FROM 16TH DAY OF GESTATION TO 21ST POSTNATAL DAY. TEN CONTROL AND HYPOTHYROID PUPS WERE INJECTED WITH 10% FORMALIN (10 ML) IN THE RIGHT HIND PAW AT 21 DAYS OF AGE. WE SCORED THE PAIN BEHAVIOR USING THE TIME-SAMPLING METHOD. IMMUNOHISTOCHEMISTRY WAS PERFORMED USING RABBIT ANTI-PERK44/42 AND SHEEP ANTI C-FOS.RESULTS: HYPOTHYROID PUPS SHOWED 50% REDUCTION IN WEIGHT GAIN AT WEANING TIME. CONTROL PUPS PRODUCED THE TYPICAL BIPHASIC NOCICEPTIVE RESPONSES IN CONTRAST TO PERSISTENCE PAIN WITHOUT ANY DISTINCT PHASE II IN HYPOTHYROID PUPS. THE SPINAL PERK IMMUNOREACTIVE NEURONS WERE FIVE TIMES MORE IN HYPOTHYROID PUPS (PHASE I) WHEREAS THE C-FOS IMMMUNOREACTIVE NEURONS WAS INCREASED IN NORMAL PUPS TWO TIMES MORE THAN HYPOTHYROID PUPS (PHASE II).CONCLUSION: THE QUANTITATIVE RELATIONSHIP OF PERK ACTIVE NEURONS IS: 3.4 FOLDS (PHASE I) MORE AND C-FOS 1.2 FOLDS LESS IN HYPOTHYROID/NORMAL PUPS (PHASE II).
