PARING OF VENTRAL TEGMENTAL AREA DOPAMINERGIC INHIBITION WITH ITS GABAERGIC EXCITATION DRIVEN BY BLOCKADE OF GABA-A RECEPTORS

Abstract

 INTRODUCTION: THE VENTRAL TEGMENTAL AREA (VTA) IS COMPOSED OF DOPAMINERGIC (DA) AND GABAERGIC NEURONS. THE EXTERNAL GABA INPUTS TO VTA HAVE REGULATORY EFFECT ON THE VTADA NEURONS BUT THE VTA-GABAERGIC IS PROPOSED AS REGULATOR ALSO. IN THIS RESEARCH THE FUNCTIONAL PAIRING OF THE VTA-DA AND GABA NEURONS WERE STUDIED BY GABA-A RECEPTOR BLOCKADE AFTER INTRACEREBROVENTRICULAR (ICV) MICRO INFUSION OF BICUCULINE.METHODS: MALE WISTAR RATS WERE PREPARED FOR VTA NEURONS RECORDING UNDER URETHANE ANESTHESIA. THE MULTIUNIT RECORDINGS COMPRISED OF VTA-DA AND GABA UNITS THAT PROVIDED BY ADJACENT MICROELECTRODE LOCALIZATION IN THE RIGHT VTA. BICUCULINE WERE MICRO INFUSED IN 3 GROUPS (5, 2.5, AND 1 MG, ICV). THE CONTROL AND VEHICLE GROUPS SHOWED NO DIFFERENCE IN PERI- STIMULUS TIME HISTOGRAM (PSTH). THE PAIRING OF THE VTA-DA, GABA NEURONS WERE EXTRACTED BY IGOR PRO 6.0 SOFTWARE OFF-LINE. THE PSTH OF SPIKES WERE ANALYZED BY ONE-WAY ANOVA WITH LSD POST HOC TEST.RESULTS: 41 VTA-DA AND 29 VTA-GABA NEURONS WERE ISOLATED WITH SIMULTANEOUS FIRING. THE BICUCULINE MICRO INFUSION EXCITED THE VTA-GABAERGIC NEURONS AND IN THE SAME TIME THE VTA-DA NEURONS WERE INHIBITED. THE DELAY TIME FOR VTA-DA INHIBITION WAS 400±53.6 MSEC AND THE DURATION OF PAIRED INHIBITION WAS 81.4±21.5 MIN DOSE DEPENDENTLY IN 83% OF RECORDINGS. THE RECURRENT OF VTA-GABAERGIC NEURONS TO BASELINE FIRING WERE COUPLED TO DECEASE OF EXCITATION OF THE VTA-DA NEURONS WITH ASYMMETRICAL MIRRORING.CONCLUSIONS: ONE OF THE MYSTERIES OF THE FUNCTION OF THE VTA IN THE PSYCHOTHERAPY, IS THE DUAL EFFECTS OF THE PSYCHOTIC AGENTS. THE MAIN IDEA ABOUT THIS PARADIGM IS THE POSTULATION OF GABA AFFERENTS TO THE VTA. THE DATA OF THIS RESEARCH REVEALED THAT ALTHOUGH THE GABA INPUTS OF THE VTA IS CRITICAL, BUT THE IN SITU VTA-GABAERGIC NEURONS PLAYS STRONG SWITCHING FACTOR FOR THE VTA-DA OUTPUT.
