Abstract

 SYNTHETIC DYES WIDELY ARE USED IN MANY INDUSTRIES. THEY ARE MAJOR SOURCES OF ENVIRONMENTAL, ESPECIALLY WATER, POLLUTION. THE EMPLOYMENT OF THESE DYES MUST BE MANAGED AND MUST BE TREATED BEFORE BEING RELEASED INTO THE ENVIRONMENT [1].ENZYMES ARE ONE OF THE BEST OPTIONS OF ORGANIC MATERIAL DEGRADATION. PEROXIDASE (POX, EC 1.11.1.7) IS A HEM-CONTAINING ENZYME THAT WIDELY THERE IS IN NATURE [2]. THIS ENZYME HAS A HIGH CAPACITY FOR OXIDATION OF PHENOLIC COMPOUNDS. IN RECENT YEARS THE USE OF PLANT TISSUE MATERIALS AND CRUDE EXTRACTS OF VARIOUS VEGETABLES AS NOVEL BIOCATALYSTS HAS ALSO RECEIVED CONSIDERABLE INTEREST FOR REPLACING ISOLATED ENZYMES.THE CRUDE EXTRACT OF KOHLRABI (BRASSICA OLERACEA GONGY LODES) IS A RICH SOURCE OF POX. ACTUALLY THIS ENZYME UTILIZES HYDROGEN PEROXIDE TO OXIDIZE A WIDE VARIETY OF ORGANIC AND INORGANIC COMPOUNDS. IN THE PRESENT WORK, EXTRACT OF KOHLRABI HAS BEEN USED AS A SOURCE OF POX FOR THE DECOLORIZATION OF ACID BLUE 113. ALSO FOR REUSABILITY AND EASILY SEPARATION OF THE ENZYME, IMMOBILIZATION OF CRUDE EXTRACT WAS DONE BY ENTRAPMENT METHOD USING SODIUM ALGINATE. PEROXIDES IMMOBILIZATION EFFICIENCY BY USING ENTRAPMENT METHOD WAS 73%.IMMOBILIZED EXTRACT CONTAINING POX WAS USED FOR ACID BLUE 113 REMOVAL FROM WATER. EFFECT OF DIFFERENT PARAMETERS INCLUDING PH, H2O2 (30%) VOLUME PER 15 MIN, DYE CONCENTRATION AND AMOUNT OF IMMOBILIZED PEROXIDASE WERE INVESTIGATED IN A STIRRED TANK BIOREACTOR. THE BEST CONDITIONS WERE OBTAINED INCLUDING; PH=7, H2O2= 0.5 ML/15 MIN, ACID BLUE 113 CONCENTRATION = 30 PPM AND AMOUNT OF 6 G IMMOBILIZED EXTRACT WHILE TEMPERATURE WAS CONSTANT AT 27°C. DYE DECOLORIZATION EFFICIENCY OF 84.5% WAS ACHIEVED UNDER OPTIMUM CONDITIONS AFTER 90MIN.
