COMPLEX FORMATION OF DESFERRITHIOCIN WITH LEAD AND BIOLOGICAL APPLICATION

Abstract

 BACKGROUND: LEAD IS A TOXIC HEAVY METAL, IT CAN CAUSE SEVERE DISRUPTION TO THE CENTRAL NERVOUS SYSTEM. DESFERRITHIONCIN (DFT) IS A TRIDENTATE BIOLOGICAL LIGAND THAT HAVE HIGH SELECTIVITY FOR BINDING PB+2 (LOGB=19.2).METHODS: IN PRESENT RESEARCH, THE TOXIC EFFECT CAUSED BY LEAD (II) IONS WERE EXPLORED BY PERFORMING IN VIVO STUDIES ON RATS FOLLOWING DRINKING ADMINISTRATION OF LEAD (II) CHLORIDE AT DOSE OF (30MG/KG BODY WEIGHT) AFTER 21 DAYS. IN ORDER TO INVESTIGATE THE ABILITIES OF DFT IN REMOVING PB+2 FROM THE RATS ORGANS THIS LIGAND WAS GIVEN ORALLY FOR ONE WEEK AND AFTER CHELATION THERAPY, ORGANS WERE SAMPLED FOR PB+2 ANALYSIS BY ICP-OES METHOD.RESULTS: A SIGNIFICANT DIFFERENCE BETWEEN CONTROL AND TREATED GROUPS WAS OBSERVED. THE GENERAL SYMPTOMS OF TOXICITY APPEARED AFTER 2 WEEKS. ABNORMAL CLINICAL SIGNS IN ANIMALS WERE APPEARED AS FOLLOWS: WEAKNESS AND LOSS OF HAIR. ALSO THE BODY WEIGHTS OF ALL ANIMALS IN TREATED GROUPS WERE SIGNIFICANTLY DECREASED. LEAD WAS FOUND IN THE KIDNEYS FOLLOWED BY INTESTINE. AFTER THE CHELATION THERAPY, THE OBTAINED RESULTS SHOWED THAT PRESENT LEAD LEVELS IN ALL TISSUES WERE SIGNIFICANTLY REDUCED AND THE SYMPTOMS ALSO DECREASED.CONCLUSION: THE RESULTS WERE SHOWED THAT THE DFT LIGAND WAS ABLE TO BIND PB+2 ION IN BIOLOGICAL SYSTEM AND REMOVE IT FROM THE BLOOD AND ORGANS.
