FACILE SYNTHESIS OF MAGNETIC MOLECULARLY IMPRINTED POLYMER FOR RAPID PRECONCENTRATION AND DETERMINATION OF PERPHENAZINE IN URINE SAMPLES

Abstract

 BACKGROUND: MAGNETIC MOLECULARLY IMPRINTED POLYMERS (MMIPS) ARE A SPECIFIC CLASS OF SOLID PHASE EXTRACTION (SPE) SORBENT THAT HAVE BOTH SELECTIVITY AND MAGNETIC PROPERTIES TOGETHER [1]. SYNTHESIS OF MMIPS ARE TIME CONSUMING AND USUALLY HAVE MANY STEPS ALONG WITH USING EXPENSIVE FUNCTIONAL MATERIAL TO PREPARE SURFACE CONTAINING REACTIVE DOUBLE BONDS [2]. IN THIS STUDIES, A NOVEL MMIPS SORBENT BASED ON MAGNETITE/METHACRYLIC ACID/POLY ((METHACRYLIC ACID-CO-ETHYLENE GLYCOL DIMETHACRYLATE) (FE3O4/MAA/P (MAA-CO-EGDMA)) WAS DEVELOPED FOR RAPID MICRO SEPARATION AND PRECONCENTRATION OF PERPHENAZINE (PPZ) IN URINE SAMPLES.METHODS: A ONE STEP SYNTHESIS WAS DESIGN TO PREPARE FE3O4 NANOPARTICLE CONTAINING REACTIVE DOUBLE BONDS. TO DO THIS, COMMON METHODS FOR THE PREPARATION OF IRON OXIDE NANOPARTICLES WERE USED, EXCEPT THAT THE ALKALINE SOLUTION WAS REPLACED BY AN ALKALINE MAA SOLUTION. FINALLY MMIPS WAS SYNTHESIZED BY DISTILLATIONPRECIPITATION POLYMERIZATION METHOD BY MIXING FE3O4@MAA, MAA, EGDMA, AZOBIS (ISOBUTYRONITRILE) AND PPZ IN ACETONITRILE WHILE HEATING AT 85OC FOR 90 MIN. EFFECT OF MMIPS AMOUNT, PH, SOLUTION VOLUME AND THE KIND OF ELUTING SOLVENTS ON PRECONCENTRATION OF PERPHENAZINE WERE INVESTIGATED. THE METHOD OF MEASUREMENT WERE HPLC-DAD.RESULTS: IN OPTIMUM CONDITION FOR PRECONCENTRATION WAS, 5 MG OF MMIPS, 5 ML OF AN AQUEOUS SAMPLE SOLUTION AT PH 4.5 AND VORTEX TIME OF 1MIN. THEN PERPHENAZINE WAS ELUTED BY 400ML OF METHANOL: ACETIC ACID (9:1). THE METHOD WAS SUCCESSFULLY APPLIED FOR THE DETERMINATION OF PERPHENAZINE IN URINE SAMPLES WITH RECOVERIES IN THE RANGE OF 89.4% TO 106.4% AND RELATIVE A STANDARD DEVIATION OF 6.8% (6 REPLICATED ANALYSIS OF PPZ). THE DETECTION LIMIT WAS 5 NG ML-1.CONCLUSION: TO OUR KNOWLEDGE, IT IS THE FIRST TIME THAT FE3O4@MAA IS PREPARED AS SURFACE CONTAINING REACTIVE DOUBLE BONDS FOR SYNTHESIS OF SURFACE MOLECULARLY IMPRINTED POLYMER FOR RAPID PRECONCENTRATION AND DETERMINATION OF PERPHENAZINE IN URINE SAMPLE. THIS METHOD ELIMINATE SILANIZATION STEP AND DOES NOT REQUIRE ADDITIONAL REAGENT FOR MODIFICATION OF SURFACE. THE RESULTS OF PARTICLE SIZE ANALYZER SHOWED THAT THE SIZES OF FE3O4@MAA NANOPARTICLES AND MMIPS WERE 22 NM AND 6 MM, RESPECTIVELY.
