Abstract

 Objective: The present study investigates the effects of ROSIGLITAZONE (a peroxisome proliferators-activated receptor gamma) on the histological parameters of the DORSAL ROOT GANGLION and its recovery potential of injured sciatic nerve in rat.Materials and Methods: Rats were divided into 4 groups including Control, sham-operated, SCIATIC NERVE CRUSHes (SNC) and SNC+ROSIGLITAZONE treatment (5mg/ kg body weight/day). Sciatic functional index (SFI) is used to evaluate functional recovery. The quantitative histological parameters were evaluated using stereological method.Results: The results revealed the volume and surface of A- and B- cells reduced in the SNC when compared with sham- operated group. After treating SNC animals with ROSIGLITAZONE the volume of A- and B-cells increased~30% and ~48% and the surface of them increased ~45% and ~21% as well as in comparison with non-treated animals after SNC. The number of the A, B- and satellite cells reduced in the SNC in comparison with sham- operated group. The number of A-, B- and satellite cells increased ~38%, ~34% and ~29% in SNC+ROSIGLITAZONE in comparison with nontreated crushed animals. The sciatic functional index (SFI) was different between sham-operated and SNC groups after nerve injury.Conclusion: SFI recovered in ROSIGLITAZONE-treated rats after SNC rats in comparison with non-treated animals.The results have demonstrated that ROSIGLITAZONE has a neuroprotective effect on the DRG and enhance the functional recovery after SNC in the rats.
