Memantine hydrochloride, 3,5-dimethyl adamantine-1-aminohydrochloride, is known as an efficient and extraordinary drug for treatment of moderate to severe Alzheimer disease. The present report includes: memantine hydrochloride introduction, study of synthetic chains, methods of synthesis in laboratory, analysis and discussion. In selected synthetic chain in this project, memantine hydrochloride was synthesized in 3 steps. In the first step, 1-bromo-3,5-dimethyladamantane was synthesized by reaction of 1,3-dimethyladamantane and bromine. In the second step, by reaction of 1-Bromo-3,5-dimethyladamantane and formamide, 1-formamido-3,5-dimethyladamantane was synthesized. Finally, in the third step memantine hydrochloride was prepared by acidic hydrolysis of 1-formamido-3,5-dimethyladamantane in hydrochloric acid(con). After recrystallization and purification, memantine hydrochloride was obtained in yield of 60% and purity of > 99% (by GC).
