Abstract: 

Anthralin, dithranol, cignolin or 1,8-dihydroxy-9-anthrone has been used clinically for about a hundred years as an antipsoriatic agent.Thorough evaluation of patients and of the relevant literature  led to the conclusion that a toxic effect of anthralin and its metabolites on the kidneys can be excluded. Anthralin was found to have no effect on keratinocyte growth except at very high doses.
First, 10-bromoanthralin was synthesized from anthralin and bromine in CS2 . Then the product was treated with various alkanthiols and thiophenol in CH2Cl2 at room temperature with stirring.  Finally, after the work up and separation by column chromatography the desired 10-alkanthioanthralins and 10-phenylthioanthralin were obtained with yields more than 70%. The chemical structure of the products were elucidated and established through their 1HNMR, 13CNMR, IR, MS spectra and microanalysis results. 
The formulations for clinical application were prepared and applied on the psoriatic patients at Manchester Skin Hospital in England.  Preliminary results were quite optimistic and no complains were made.  These formulations showed less side effects which are associated with other anthralin derivatives i.e. erythema at the site of application and the staining of the skin to a purple colour.
Preliminary results have shown that the two side effects were reduced while the efficacy of the drugs remained the same.  No toxicity effects were observed or reported.  The results are promising but more works are needed to have a better understanding and a clear view of the drugs and especially about their toxicity effects.
