IS IT POSSIBLE TO PREDICTE P53 GENE CHANGES WITH SERUM P53 AUTOANTIBODY TESTINGIN PATIENT WITH BLADDER TRANSITIONAL CELL CARCINOMA?

Abstract: 

PROPOSE: MUTATION OF P53 GENE WERE DEMONSTRATED IN MANY SOLID TUMOR WITH VARYING FREQUENCY. WE ANALYSED THE RELATIONSHIP BETWEEN P53 AUTOBODY IN SERUM, P53 PROTEIN OVER EXPRESSION IN BLADDER TUMOR TISSUE, AND PATHOLOGIC GRADING OF BLADDER TUMORS OF 50 PATIENTS WITH AND 15 PATIENTS WITHOUT BLADDER TUMOR.
METHODS & MATERIALS: IN 50 PATIENTS STUDIED, BLADDER CANCER WAS DIAGNOSED AS GRADE 1 IN 12 CASES, GRADE 2 IN 30 AND GRADE 3 IN 8 PATIENTS. ANTI P53 ANTIBODY WERE DETECTED BY AN ENZYME LINKED IMMUNOSORBENT ASSAY. IMMUNOHISTOCHEMICAL STAINING WAS PERFORMED USING CATALYSED SIGNAL AMPLIFICATION (CSA) SYSTEM.
RESULTS: WE FOUND 10% P53 AUTOANTIBODY-POSITIVE SERA WITHOUT A STATISTICALLY SIGNIFICANT CORRELATION WITH TUMOR GRADE. OF 41 PATIENTS, 11 SHOWED P53 OVER EXPRESSION IN IMMUNOHISTOCHEMICAL STAINING. ALL OF SERA POSITIVE PATIENTS HAD P53 PROTEIN-POSITIVE TUMORS. CONTROL SERA WERE ALL NEGATIVE FOR P53 AUTOANTIBODY.
DISCUSSION & CONCLUSION: THIS STUDY SHOWS A STRONG RELATIONSHIP BETWEEN P PROTEIN OVER EXPRESSION AND THE OCCURRENCE OF P53 AUTOANTIBODY IN BLADDER CANCER. BUT THIS ELISA SEROLOGIC TEST HAVE LIMITED VALUE IN PREDICTING THE ALTERNATION IN P53 GENE, BECAUSE IT HAVE HIGH RATE OF FALSE NEGATIVE RESULTS.
