Abstract: 

INTRODUCTION: AS THE FIRST ATTEMPT IN OUR COUNTRY TOWARDS PRODUCTION AND IMPLEMENTATION OF BLADDER WALL ACELLULAR MATRIX GRAFT, THE CURRENT STUDY WAS CARRIED OUT TO PRODUCE THE RAT BLADDER WALL ACELLULAR MATRIX.
MATERIALS AND METHODS: AN ADULT, MALE, HEALTHY RAT UNDERWENT SUBTOTAL CYSTECTOMY. THE MUSOCA OF THE HARVESTED BLADDER WAS SCRAPED OFF WITH A PAIR OF GLASS SLIDES. THE BLADDER WAS THEN TREATED WITH SODIUM AZIDE, DNASE, AND SODIUM DESOXYCHOLATE IN A THREE-STEP PROCESS, 4-6 HOURS EACH.
RESULTS: LIGHT MICROSCOPY FOLLOWING H & E AND TRICHROME STAINING REVEALED CELL-FREE EXTRACELLULAR MATRIX, WITH INTACT COLLAGEN BUNDLES.
CONSLUSION: RAT BLADDER CAN BE MADE ACELLULAR THROUGH STANDARD THREE-STEP PROTOCOL WITH CELL LYSIS, ENZYMATIC EXTRACTION OF THE GENETIC MATERIAL, AND WASH AWAY OF THE LIPID MEMBRANES.
