Abstract: 

Objective:  Peroxisome Proliferator Activated Receptor γ2 (PPARγ2) is a nuclear receptor that regulates adipocyte differentiation, lipid metabolism and insulin sensitivity. The aim of this study was to evaluate effect of the Ala12 Allele of PPARγ2 gene on BMI in Iranian Diabetic and Obese Subjects.
Methods: The genomic DNA’s of the 312 subjects including type II diabetes and obesity were extracted. Pro12Ala polymorphism was detected by Polymerase Chain Reaction–Restriction Fragment Length Polymorphism (PCR-RFLP) analysis.
Results:  Frequencies of Ala in obese subjects (q Ala=0.166) were significantly different from those control subjects (q Ala = 0.089) OR95%CI 2.358 (1.101-5.05) (pvalue=0.025). In contrast, no significant association was seen between the Pro12Ala polymorphism and type II diabetes OR 95%CI 0.652 (0.261-1.628). In all subjects, the Ala carriers had higher BMI compared to the common allele.
Conclusion: Our results showed that Pro12Ala polymorphism in PPARγ2 gene is associated with obesity in our Iranian subjects and presence of the Ala allele associated with higher BMI.
