Abstract: 

Introduction: Beta-adrenergic antagonists have been extensively used in treatment of several cardiac diseases such as hypertension, cardiac arrhythmias, congestive heart failure and angina pectoris. The inhibitory effect of Propranolol on human lung tumor cell proliferation, mesovarian leiomyomas development in rat and its cytotoxic effect on lung adenocarcinoma have been reported. The cytotoxic effect of Propranolol on some tumor cells as well as its extensive and continuing use in a number of heart diseases incited us to examine its cytotoxicity on a fibrosarcoma WEHI-164 cell line.
Methods: The fibrosarcoma WEHI-164 cells were incubated with different concentrations of Propranolol (0.1 -100 µg/ml) prepared in complete RPMI cell culture medium for 6, 12, 24 and 48 hours. The cytotoxicity was assessed by 3-[4,5-dimethyl thiazol-2,5-diphenyltetrazoliumbromide] (MTT) reduction and trypan blue dye exclusion methods.
Results: Propranolol caused a significant concentration dependent cytotoxic effect on mouse fibrosarcoma WEHI-164 cell line after 6 hours compared with untreated control cells.
Conclusion: Our results showed that fibrosarcoma WEHI-164 cell line was dose dependently sensitive to Propranolol. These findings suggest further studies for evaluating anti-tumoral effect of Propranolol.
