Abstract: 

Scopolamine is a drug with alkaloid and anti cholinergic properties. Scopolamine is a muscarinic antagonist that affinity to receptor. On the other hand physostigmine is an inhibitor for the cholinesterase enzyme. Physostigmine directly stimulates nicotinic and muscarinic receptors by interfering acetylcholine metabolism. Opioids are one of the drugs that reduce C.N.S activity. In present research the effect of improvement and deterioration of memory on creation morphine dependence in male mouse by CPP (conditioned place preference) method had investigated. Scopolamine had used as weakling agent of memory in 2 dose and physostigmine had used as improving agent of memory in 0.1 dose. Morphine dose had used in 10 dose. There are three stages in CPP method preconditioning, conditioning, post conditioning. The CPP test had done in a box with two equal parts. One of them was black and the other is white. Animals weight was between 25-35gr and they were divided in to 8 group with 12 member. The preconditioning stage is one day and each mouse separately puts in the box and can freely access to both parts for 10 minutes. In conditioning stage the control group had not received any drug and sham group had received only water .Other experimental groups had received its special drug. In this stage drugs were injected subcutaneously at fixed time once a day for 3 to 8 days. Each mouse randomly puts in white or black part for 30 minutes every day. In post conditioning stage each mouse put in CPP box for 10 minutes and access to hole box. Average time for each mouse had recorded in part that coupled with drug. Statistical analysis had shown average time that spending in drugs coupled part in different groups had depended to the type of the drug that used. At final evidences had shown that improving memory with physostigmine lead to increase creation of morphine dependence. On the other hand deterioration memories with scopolamine lead to decrease creation of morphine dependence. Simultaneously of these two drugs lead to neutralize each other and the conclusion is the same as the morphine group.
