THE EFFECTS OF BISPHENOL A, ALONE OR IN COMBINATION WITH PROCYANIDIN A2 ON GLUCOSE HOMEOSTASIS AND PDX1, GLUT2 GENE EXPRESSION IN MALE MICE

Abstract

 INTRODUCTION: PROCYANIDINS ARE ABUNDANT IN PEOPLE’S DIET AND HAS ANTIDIABETIC PROPERTIES. BISPHENOL A (BPA) IS A COMMON CHEMICAL MATERIAL USED IN FOOD AND BEVERAGE PACKAGING. EXPOSURE TO BPA CAN BE INVOLVED IN CAUSES OF TYPE2DIABETES. THE MAIN OBJECTIVE WAS TO EXPLORE THE EFFECT OF PROCYANIDINA2 ON RESOLVING BPA SIDE EFFECTS ON THE SERUM LEVEL OF GLUCOSE AND INSULIN, EXPRESSION OF PDX1AND GLUT2 GENES. ANOTHER AIM WAS APOPTOSIS EXAMINATION IN THE ISLET CELLS.METHODS: IN THIS STUDY FIRST THE APPLICATION OF FIVE DOSES OF PCA2 (3, 10, 30,100 AND 300 MM) WAS EXAMINED, ALONE AND WITH100MG/L BPA, ON THE INSULIN SECRETION FROM ISOLATED ISLETS OF MICE AT IN VITRO CONDITION. NEXT THE IMPACT OF PCA2 ON ISLET CELLS APOPTOSIS WAS EVALUATED. AFTER THIS, AT IN VIVO CONDITION THE BPA AND PCA2 ADMINISTRATION WAS PERFORMED FOR20DAYS.NEXT BLOOD GLUCOSE AND INSULIN, PDX1AND GLUT2 GENE EXPRESSION WERE TESTED.RESULTS: OUR FINDING INTRODUCED THE10ΜM CONCENTRATION OF PCA2 AS AN EFFECTIVE DOSE FOR INSULIN SECRETION. IN VIVO EXPERIMENTS INDICATED THAT BPA INCREASES FASTING BLOOD GLUCOSE LEVEL, BUT THE CO-ADMINISTRATION OF PCA2AND BPA MODIFIES IT. REAL TIME PCR RESULTS SHOWED THAT BPA REDUCES GENE EXPRESSION OF PDX1AND GLUT2IN PANCREAS WHEREAS THE COADMINISTRATION OF BPA AND PCA2 IMPROVES IT. FINALLY APOPTOSIS ASSESSMENT USING FLOWCYTOMETRY INDICATED THAT BPA INDUCES APOPTOSIS IN ISLET CELLS AND PCA2 STRONGLY PREVENTS SUCH PHENOMENA.CONCLUSION: WE PROVIDED WITNESSES EXPRESSING THAT DECREASING OF APOPTOSIS IN ISLET CELLS, OVER EXPRESSION OF PDX1 AND GLUT2GENES IN PANCREAS TISSUE ARE HYPOTHETICAL MECHANISMS BY WHICH PCA2PREVENTS HYPERGLYCEMIA.
