Abstract: 

Recent studies indicate that Artemisia extract and its active substance, artemisinin, is effective against coccidiosis. However, there is no report about the mechanism of drug in treatment of coccidiosis. The aim of this study was to investigate the location of the drug action in the intestine of coccidiosis infected chickens using immunohistochemistry (IHC) technique.
Artemisinin was derivatizied to the dihydroartemisinin carboxymethylether containing a reactive carboxyl group, and then conjugated with BSA by a mixed anhydride method. Immunization and the production of antibody were conducted by injection of artemisinin-BSA conjugate in rabbit. Immuno-blot staining confirmed the presence of antibody in immunized serum. The antibody was concentrated with saturated ammonium sulfate and then labeled with FITC. Chickens were experimentally infected to coccidiosis by administration of 250000oocycts/chick. The infected chicks were treated with a granule containing artemisinin and pure artemisinin for 5 days. The chickens were killed and the tissue samples were collected from intestines and cecum, and they were used for IHC.
The percent of staining antrocytes cells in the crypt and villi epithelium was used as the main criteria for the severity and grading of artemisinin staining. It was found that many artemisininstaining in the epithelium cells of the cecum of the tissue taken from the chicken treated with either granule containing artemisinin or pure artemisinin.
The study demonstrated the effectiveness of the artemisinin in the location of Eimeria tenella invasion and the protective effect of prepared artemisinin granule in experimental coccidiosis in broilers. This can be a preliminary evidence for understanding the pharmacodynamics of artemisinin in coccidiosis.
