The Punicaceae plant, Punica granatum LINN, is distributed throughout the Mediterranean, eastward to China and India. The flower parts serve as a remedy for diabetes mellitus and the methanolic extract of the flower of P.granatum was found to inhibit tumor necrosis factor-a (TNF-a). The aim of the present study was to investigate the effect of P.granatum flower hydroalcoholic extract on the uterus of rats. Extract was provided. Uterus of rats was suspended in an organ bath containing Dejalon at 29oC. Tissue contractility was isometrically recorded. Potassium chloride (60mM), BaCl2 (4mM), propranolol (1μM) and oxytocin (10mU/ml) were applied to the tissue in the presence or absence of the extract. Application of the extract (p<0.05) before oxytocin reduced the contractions amplitude of oxytocin in a dose dependent fashion. The extract added at 0.5, 1, 2, 4 mg/ml cumulatively to the organ bath reduced this contraction but had no effect on uterus contraction induced by BaCl2. Spasmolytic effect of extract was not affected by propranolol (1μM for 5 min) in KCl-induced contractions. The extract blocked voltage-dependent calcium channel but it did not have any effect on b-adrenoreceptors and potassium channels.
