Abstract: 

Seminal plasma and spermatozoa have antioxidant capacity for neutralization of reactive oxygen species (ROS). Higher ROS generation and lower antioxidant capacity was reported in teratozoospermia compared to normozoospermia. We evaluated the effect of an antioxidant, a-tocopherol, on teratozoosperm motility, viability, and acrosome reaction (AR) in control condition and when AR-induced by calcium ionophore A23187. Teratozoospermic semen samples were obtained from 15 volunteers 20-30 years old after 3-5 days of sexual abstinence. Samples were washed, centrifuged and incubated in 37°C and 5% CO2 until sperms swimmed-up. Sperms were counted in the supernant and divided into five groups, each contained 2×106 sperm/ml. Groups 1 to 5, were incubated for 1 hour with Ham’s F-10 solution as control group, 1% of ethanol, 10μM A23187, 40μM a-tocopherol, 10μM A23187+40μM a-tocopherol, respectively. Sperm motility was assessed by light microscopy, sperm vitality determined by eosin staining, and AR evaluated by FITC-PSA staining. a-tocopherol induced a significant improvement in percent of motile and live sperms as compared to control group. Whereas A23187 had the opposite effect and reduced the percent of motile and live sperms. On the other hand, incubation of sperms in a medium containing A23187 and a-tocopherol showed that a-tocopherol has a potency to reverse the adverse effects of A23187. A23187 induced AR in teratozoosperms, while a-tocopherol significantly diminished this effect. In conclusion, a-tocopherol has a potency to improve the biological functions of teratozoosperms. It seems that the harmful effects of A23187 are due to excessive ROS production and a-tocopherol neutralizes these effects.
