Abstract: 

The objective of this study was to assess the effects of LHRH-A2 (a gonadotropin releasing hormone analog), pimozide and chlorpromazine (dopamine receptor antagonists), alone or in combination, on induction of maturation and spawning of female brood stocks of Kutum. 81 female brood stocks of Kutum were studied in Shahid Ansari Fish Rearing, Propagation and Restocking Center during reproductive season (March and April 2007). Treatments included: 1. Single injection of 1 mg/kg of CPE as a positive control, 2. LHRH-A2 (1 μg/kg), 3. pimozide (5 mg/kg), 4. LHRH-A2 + pimozide (1 μg/kg + 5 mg/kg), 5. chlorpromazin (2.5 mg/kg), 6. LHRH A2 + chlorpromazine (1 μg/kg +2.5 mg/kg), 7. LHRH-A2 + pimozide + chlorpromazine (1 μg/kg + 5 mg/kg +2.5 mg/kg), 8. negative control group receiving 0.7% saline (1 ml). A group of normal female brood stocks was also included that received no injection. For histological studies, ovary samples of non-ovulated females were fixed in bouin; the routine proceduresof preparation of tissues were performed and the paraffin blocks were cut at 5-7 microns, stained with H&E and studied under light microscope. The results showed that the highest percentage of ovulated females belonged to treatment groups 1 and 4 (77.78%). Minimal percentage of ovulated females (22.22%) and maximum fertilization success (98.1± 0.21 a) belonged to treatment group 5. Group 6 showed the lowest mean latency period (31.83± 8.3a). Maximum ovulation index (84.2± 2.82 a) belonged to treatment group 7. Non-ovulated females were in negative control and normal groups. The microscopic observations of ovary in non-ovulatedbrood stocks showed that the oocytes in these treatments were maturated, being in the final phase of stage IV.
