Abstract: 

Recent experimental and clinical data suggest that c-peptide replacement during type 1 diabetes exerts beneficial effects on diabetic neuropathy. The aim of this study was to determine if physiological dose of c-peptide administration had beneficial effects on metabolic status and renal function type 1 diabetic rats. 3 groups of rats were used for this study: 1) non-diabetic group 2) diabetic group 3) diabetic group that received c-peptide (25nmol/kg/ip/day). Streptozotocin was used for diabetes induction (60 mg/kg/iv). Weight was assessed every week. Blood and urine glucose levels were measured with glucose stripe test (Accucheck), and urine protein assessed with stripes. Despite 9% increase in weight in non-diabetic group, 18% and 15% weight loss was seen in diabetic and diabetic groups that received c-peptide, respectively. C-peptide replacement could improve weight gain in diabetic group that received c-peptide. Glucosurea & proteinurea was not seen in non diabetic group, but marked glucosurea and proteinurea were seen in diabetic group (P<0.05). In diabetic group that received c-peptide only proteinurea diminished significantly (P<0.05) and reduction in glocusurea was not significant. C-peptide administration to streptozotocin diabetic rats induces weight gain regardless of hyperglycaemia or glucosurea. Rats supplemented with c-peptide exhibit better renal function in comparison to diabetic group.
