Abstract:

Punica granatum L. flowers, rich in tannins and poly-phenolic compounds, have anti-inflammatory, anti-oxidant and mucosal protective properties which make it a good candidate for inflammatory bowel diseases therapy.
Hydroalcoholic (0.5, 1.0, 1.5 g/kg/d) and buthanol extract (125, 250, 375 mg/kg/d) of the plant flowers were administered to separate groups of male Wistar rats (n=6) both orally and intra-rectally for 5 days. Control groups received normal saline (2 ml/kg/d), prednisolon (4mg/kg/d), asacol enema (400 mg/kg/d) and hydrocortisone enema (20 mg/kg/d). Colitis was induced by using 2 ml of acetic acid 4% via rectum. All rats were sacrificed 24 hours later and the tissue injures were assessed macroscopically and histopathologically.
Two mentioned extracts with all doses used were effective to reducecolon wet weight/ length ratio. Higher oral and rectal doses were also effective to decrease ulcer severity, area and index as well as inflammation severity and extent, and total colitis index compared to control groups.
It is concluded that Punica granatum L. flowers were effective to protect against experimental acute colitis in such a manner that was dosedependent and independent to the route of administration. Since the buthanol extract was equally effective while it was used with 4 fold smaller dosage range, it is assumed that the poly-phenolic compounds play an important role in our findings.
