Abstract: 

In this study the oxidative stress of diazinon (DZ) and B6 protective effect against DZ in astrocytes typs U373MG were evaluated by determination of GSH level and cells viability.
Material and methods: The cells were exposed to the different doses (1, 10, 30, 100, 300 μM) of DZ for different time intervals including, 24, 48 and 72 hours. Then, the dose of 300 μM of DZ for 72 hrs determined as optimum dose. Then the effects of DZ on GSH level and cells viability in the presense of B6 as either pre or post treatment was evaluated.
Results: Although the dose of 100 μM of DZ increased slightly, but the dose of 300 μM significantly reduced the GSH level at 72 h post exposure. Pre treatment with B6 30 min before DZ could increase the GSH level approximately near the control value. Also, B6 alone increased significantly GSH level. DZ could decrease cells viability significantly (about 20%) after 72 hs. B6 pre or post treatment could prevent cell death induced by DZ.
Conclusions: these findings indicated that DZ is toxic for U373MG and decreases GSH level by oxidative stress induction. Either pre or post treatment by B6 can protect cells and prevent GSH level reduction and cell death.
