Abstract: 

The nucleus paragigantocellularis latralis (LPGi) is located in the rostral ventrolateral medulla (RVLM), a brain stem region that regulates homeostatic functions such as blood pressure and cardiovascular reflexes, respiration, pain and sexual behavior. In this research, the effects of clonidine on locomotor activity in LPGi lesioned and intact male rats were studied. Animals were divided into 2 groups and each group had 3 subgroups: Control (intact or lesiond) and two groups that they received clonidine 0.02 mg/kg and 0.2 mg/kg, respectively. In lesiond group Animals were anesthetized with ketamine (110 mg / kg) and rampone (3 mg / kg) mixture (i.p.); Stainless style electrode were placed in LPGi (AP = 11.8, L= ± 1.6, DP = 10.5) bilaterally. LPGi was lesioned by 1Ma electrical current for 6 sec. After recovery period loco motor activity were tested by hand method's cage. Number of entrance to each box in 10 minutes was considered as loco motor index. 0.02 mg/kg (P<0.048) and 0.2 mg/kg (P<0.001) clonidine decreased significantly locomotor activity in lesiond group but did not have any effect in intact group. Locus coeruleus (LC) is important adrenergic nucleus in brain that receivedexcitatory projection from LPGi. It seems that after LPGi lesion, alpha-2 adrenergic receptors up regulation were occurred and these receptors accelerated clonidine action in decreasing of noradrenalin release or decreasing of electrical activity of LC or other nucleus that has important role in locomotion activity.
