Abstract: 

Endothelin-1 (ET-1) is a potent vasoconstrictor peptide produced by vascular endothelial cells. In the lung, highest levels of ET-1 are secreted by endothelium, smooth muscle airway epithelium and a variety of other cells. Exercise is an important factor that affects ET-1 expression and production. This substance plays an important role in many physiological and pathological conditions as well as in the function of lung. In the present study the effects of six months exercise training on the expression of ET-1 gene in lungs tissue were investigated. For this purpose, 20 male Wistar rats (235 ± 27) were selected. Theywere maintained on a 12:12-h light-dark cycle and received food and water. On the day of the experiment, rats were randomly divided into two groups (n=10). They ran on a treadmill for 60 min at a speed of 25 m/min daily for 6 months. 48 h after this exercise the lungs were removed and were stored in -70°C. Total RNA was extracted from the lung tissue and the expression of ET-1 mRNA was assessed by RT-PCR. The expression of ET-1 mRNA in the lung was significantly higher in the exercised rats than in the control ones (P<0.05). It is concluded that six months exercise increased the ET-1 expression in the lung. However, there is still need to investigate whether this change could increase or decrease its blood flow, as well as the long exercise-induced changes in ET-1 expression are needed to be investigated.
