Abstract: 

Nalidixic acid is used as an antimicrobial agent from 1962. Several methods have been established for detection and determination of nalidixic acid. High performance liquid chromatography is used as one of the best methods for analysis of this compound. In HPLC method ultraviolet and fluorescence detection revealed as suitable detectors. In this study we compared UV and fluorescence detection of nalidixic acid in reverse phase chromatography (RP-HPLC).
Method; Instruments; HPLC younglin 930D, waters binary younglinn pump, Nova pack C18 column (3.9 mm, 250 mm), fluorescence Grotom FD500, waters UV detector 450, Autochro win software program. Acetonitrile / Ammonium acetate (50/50) as mobile phase with pH=3 (adjusted by ortophosphoric acid), Ciprofloxacin (1 mg/mL) as internal standard StocR Solution for UV detection and Propylparaben (1 mg/mL) as internal standard Stock Solution for fluorescence detection.
Results; 1- Both detectors have shown specificity and selectivity.
2- Linearity for UV detection was broad (50
mg/mL – 50 ng/mL) but in low concentrations <50 ng/mL fluorescence detector have shown suitable reproducibility and repeatability.
We conclude that UV detection is better for samples with >50 ng/mL and fluorescence detection for <50 ng/mL of nalidixic acid.
