Abstract: 

Piroxicam is a poorly water-soluble anti-inflammatory and analgesic drug. There are several techniques to enhance of poorly soluble substance. Among them, the technique of liquisolid compacts is a promising technique to increase dissolution rate of poorly soluble drugs. The aim of this study was to assess the use of liquisolid technique in enhancing dissolution rate of piroxicam for improvement analgesic effect in in-vivo study. Study was performed in eleven groups of male rats. The powder, current capsule available in Iran pharmacy market and prepared liquisolid formulation in doses 2, 5, 10 mg/kg were given orally , and one hour later, formalin 2.5% (v/v) was injected to the rat, and their tolerance to pain were recorded in time intervals of 0-5 min(acute phase) and 15-60 min (chronic phase).
In chronic phase analgesic effect of liquisolid formulation of piroxicam more analgesic effects was observed more than powder and current capsule which is available in Iran pharmacy market, however in acute phase there was not any significant different. The liquisolid formulation of piroxicam induced greater analgesic effect during chronic phase of formalin test when compared with pure powder and capsule powder (p<0.001).
We can be concluded that liquisolid technique could be used for increasing the analgesic effect of piroxicam as a poorly water soluble drug.
