Abstract: 

Tamoxifen is a nonsteroidal antiestrogen which is prescribed for treatment of breast cancer. In the major aim of this study is to investigate the effect of Tamoxifen on the histological structure of uterus estrogen, progesterone and LH concentration in the female Wistar rats.
Three groups of rats (200-250gr range) gavaged with doses of 200, 400 and 600ug/kg B.W Tamoxifen dissolved in solvent (ethanol 60% and physiological solution) for 30 days. The sham group was gavaged with solvent, and control did not receive durg. On days 1 and 60 after the end of treatment, blood serum of hormones concentration was measured with RIA method; also sections of uterus stained with H&E were investigated histological. Results of the observation were analyzed by the ANOVA oneway and using SPSS software determined ±SE the means Result were termed significant if P 0.05. In the first sampling estrogen, progesterone, LH concentration, the thickness of uterus layers (perimetrium, myometrium and endometrium) and the number gland of endometrium in experimental groups were decreased in compared with control (P<0.001).
In the second sampling the estrogen, progesterone, LH concentration, thickness of uterus layers and the number gland of endometrium in experimental groups, increased compared with first sampling.
These finding suggest that Tamoxifen decreases the fertilization ability and has negative effects uterus tissue and hormones in female Wistar rats. More of negative effect of Tamoxifen on reproduction ability, is abolished with time elapsed.
