Abstract: 

THE AFLATOXINS ARE A GROUP OF STRUCTURALLY RELATED TOXIC COMPOUNDS PRODUCED BY CERTAIN STRAIN OF THE FUNGI ASPERIGILLUS FLAVUS AND A. PERASITICUS UNDER FAVORABLE CONDITIONS OF TEMPERATURE AND HUMIDITY. THE MOST PRONOUNCED CONTAMINATION HAS BEEN ENCOUNTERED IN TREE NUTS, PEANUTS, AND OTHER OIL SEEDS, INCLUDING CORN AND COTTONSEED. THE BROAD ISTRIBUTION OF THESE TOXINS IN NATURE ESPECIALLY IN FOODSTUFF AND THEIR LONG STABILITY IN FOODSTUFF IS A DANGER BOTH FOR HUMANS AND ANIMALS. IN THE PRESENT STUDY, THE NEUTRAL RED CYTOTOXICITY ASSAY WAS USED TO DETERMINE THE CYTOTOXIC EFFECTS OF AFLATOXINS (B1, B2, G1, G2) IN HUMAN HEPATOMA (HEPG2) CELL LINE. BRIEFLY, 25,000 CELLS WERE SEEDED IN EACH WELL OF A 96-WELL TISSUE CULTURE PLATE AND INCUBATED FOR 24 H, IN A HUMIDIFIED CO2 INCUBATOR AT 37°C. CULTURE SUPERNATANT FROM EACH WELL WAS THEN REPLACED WITH 200UL OF FRESH CULTURE MEDIUM CONTAINING VARIOUS CONCENTRACTIONS (2.5 5, 10, 20, 30, 40, 60, 80, 120, 160NG/ML) OF AFLATOXINS (B1, B2, G1, G2) WERE EVALUATED IN HUMAN HEPATOMA (HEPG2) CELL LINE. AFTE3 H INCUBATION AT 37°C, VIABLE CELLS WAS EVALUATED.  THE CELLS WERE WASHED AND FIXED WITH CACL2 (1%) IN HCHO (0.5%) AND FINALLY LYSED WITH CH3COOH (1%) IN C2H5OH (50%) TO EXTRACT THE DYE FROM VIABLE CELLS. THE OPTICAL DENSITY (OD) OF EXTRACTED DYE WAS THEN MEASURED WITH A VISIBLE SPECTROPHOTOMETER AT 540 NM. OUR RESULTS INDICATED THE LC50 OF B2>B1>G1> G2 (15, 16.9 21.7 AND 37.4 NG/ML RESPECTIVELY).
