Abstract: 

CISPLATIN IS ONE OF THE MOST USEFUL ANTICANCER DRUGS, FOR THE TREATMENT OF MALIGNANT TUMORS SUCH AS LUNG, HEAD AND NECK AND SPECIALLY THE OVARIAN CARCINOMA. CP FORMS DIFFERENT METABOLITE IN VIVO AND IN VITRO. IN THIS STUDY THE CYTOTOXIC EFFECT OF CP AND TWO OF ITS METABOLITES MP (MONO METHIONIN) AND BP (BIS METHIONIN) WAS MEASURED IN SKOV3 AND A549 CELL LINES. SKOV3 AND A549 CELLS WERE GROWN IN PETRIDISH AND MAINTAINED AT 37˚C IN 5% CO2 INCUBATOR. CELLS WERE EXPOSED TO 0-33.3µMOL OF CP, MP AND BP IN LOGARITHMIC PHASE FOR 1 HOUR .THEN THE CELLS WERE SEEDED IN 6 WELL PLATES FOR COLOGENIC ASSAY. COLONY COUNT WAS PERFORMED AFTER 10-12 DAYS IN DRY FREE-FRESH MEDIA.PERCENTAGE OF COLONIES FORMED AFTER EXPOSURE TO THE DIFFERENT CONCENTRATIONS OF DRUG COMPARED TO THE CONTROL WAS MEASURED AS THE CYTOTOXIC EFFECT OF THE DIFFERENT AGENTS. IC50 OF CP, MP AND BP IN THE LUNG CELL LINES WERE CALCULATED AS 10.5µM, 32 µM AND >33.3µ MAND IN THE OVARIAN CELL LINE AS 3.33µM, 23µM AND >33.3 µM, RESPECTIVELY. DOSE MODIFICATION FACTOR (DMF) FOR CP COMPARE TO MP WERE CALCULATED AS EQUAL TO 0.7 FOR LUNG CELLS AND 0.29 FOR THE OVARIAN CELL LINE. OUR RESULTS CONFIRMED THAT CP HAS A HIGHER CYTOTOXICITY (ABOUT 3 TIMES) IN OVARIAN VS LUNG CELLS ,WHICH IS ANYWAY MORE THAN ITS METABOLITES ON ANY OF THESE CELL LINES ,MP SHOWED A LOWER CYTOTOXIC EFFECTS THAN BP.DMF CALCULATION CONFIRMED THAT CP CYTOTOXICITY WAS DECREASED BY METHIONIN 2.4 TIMES LESS THAN OVARIAN CELLS COMPARED TO WHAT WAS RESULTED FROM THE LUNG CELL LINES.
