MOLECULARLY IMPRINTED POLYMER LAYER-COATED SILICA NANOPARTICLES TOWARD SOLID PHASE EXTRACTION OF GALLIC ACID FROM GREEN TEA SAMPLES

Abstract

 GALLIC ACID (3,4,5-TRIHYDROXYBENZOIC ACID) IS A NATURALLY OCCURRING PLANT PHENOL WHICH IS DERIVED FROM CERTAIN TANNINS, GALLNUTS, LEMONS AND GREEN TEA LEAVES [1]. GALLIC ACID SHOWED SELECTIVE CYTOTOXICITY AGAINST A VARIETY OF TUMOUR CELLS, WITHOUT HARMING HEALTHY CELLS [2]. IN THIS PAPER, THE MOLECULARLY IMPRINTED POLYMER (MIP) LAYER OF GALLIC ACID AT THE SILICA NANOPARTICLES SURFACE WITH SELECTIVE RECOGNITION HAD BEEN CONSTRUCTED. THE METHACRYL GROUPS OF FUNCTIONAL MONOMER AT THE SILICA NANOPARTICLES SURFACE WERE MADE REACTIVE SITES TO INDUCE IMPRINTING POLYMERISATION. THE RESULTS SHOWED THAT THE MOLECULARLY IMPRINTED POLYMERS MOREOVER HIGHLY ABSORPTION CAPACITY AND RECOGNITION SELECTIVITY, HAD AN EXCELLENT AFFINITY TOWARD GALLIC ACID. THE MIP WAS SUCCESSFULLY EMPLOYED AS SOLID PHASE IN SPE PROCEDURE COUPLED WITH HIGH PERFORMANCE LIQUID CHROMATOGRAPHY FOR DETERMINATION OF GALLIC ACID. UNDER OPTIMIZED CONDITIONS, THE SPIKED SAMPLES WERE ANALYZED WITH ACCEPTABLE HIGH RECOVERIES (96%). FOR THE PROPOSED MISPEHPLC-UV METHOD, THE LINEARITY BETWEEN RESPONSES AND CONCENTRATIONS OF GALLIC ACID WAS FOUND OVER THE RANGE OF 0.1-3000, WITH A LINEAR REGRESSION COEFFICIENT OF 0.999. THE LIMIT OF DETECTION (LOD) AND LIMIT OF QUANTIFICATION (LOQ) BASED ON THREE AND TEN TIMES OF THE NOISE OF HPLC PROFILE WERE 0.026 AND 0.087 MG.L-1, RESPECTIVELY. THE RELATIVE STANDARD DEVIATION (RSD) FOR FIVE REPLICATE MEASUREMENTS WAS 2.57%.
