SEPARATION OF X AND V-BEARING MOUSE SPERMATOZOAE BY PURE SPERM GRADIENT METHOD

Abstract: 
Objective. Sex preselection is a good idea for reduction of X-linked genetic disease. so, this study was started to evaluate the efficiency of Puresperm to sparate X-and Y -bearing sperms in mice.
Materials and Methods. Epidydimal semen centrifuged through 8 gradients of Puresperm ranged from 35% to 84% (with a 7% difference). Sperms resulting from two layers of Pure sperm (35% and 84%) were used for mouse IVF. Then 2-4 cell embryos were analyzed by multiplex PCR for Sry gene.
Results. The percentage of X-bearing sperms in 35% Pure sperm and control group (without sperm processing) was 74% and 52.8% respectively (P<0.05) and the percentage of X-bearing sperms in 84%. Pure sperm and control group was 80% and 47.2% (P<0.05) respectively.
Conclusion. SRY-PCR showed that Pure sperm discontinuous gradient enriched Y -bearing sperms in 35% layer and X-bearing sperms in 84% layer.
