IN VITRO GROWTH INHIBITION OF K562 CELL LINE WITH CELL WALL PROTEIN FRACTION OF LACTOBACILLUS CASEI AND LACTOBACILLUS PARACASEI

Abstract: 

Background: The goal of this study was to evaluate the effect of high and low molecular weight of protein fraction obtained from Lactobacillus casei and L. paracasei cell wall on K562 cell line.
Methods: Bacteria cultured in special medium and incubated in anaerobic condition and washed with PBS and then sonicated and centrifuged for cell wall separation. Protein precipitation was conducted with trichloroacetc acid (72%) and sodium de oxalate (15%). Then different concentrations (500, 1000, 2000, 3000 and 4000
mg/ml) of cell wall proteins were prepared in cell culture medium for each bacteria separately. In vitro anti cancer properties of both bacteria proteins were assayed in 12, 24, 48 and 72 hour with a colorimetric method (MTT) with 10000 cells per well.
Findings: Results showed that high and low molecular weight proteins of both bacteria could significant (p
=0.046) inhibit the growth of K562.
Conclusion: Based on the results we conclude that high molecular weight of both probiotic bacteria significantly could inhibit the growth of K562 than the low molecular protein fractions. Also, protein fraction of L. paracasei significantly had anti cancer properties than protein fraction of L. casei.
