Abstract: 

Background: There are some reports on reduction of aneuploidy rate following longer co-culture of human blastocysts. Present study aimed to evaluate post-fertilization culture until 6 and 7 days and hatching stage in self-correction of aneuploidies.
Materials and Methods: Thirty- three surplus embryos of PGD candidates were reanalyzed by fluorescence in situ hybridization in days 6 or 7 after fertilization randomly. Several cells of each embryo with good signals were studied for 8 chromosomes including 13, 15, 18, 19, 21, 22, X and Y chromosomes.
Results: Totally 302 nuclei were analyzed in 33 embryos with an average of 9 nuclei per embryo. Mosaicism were observed in 28 (85%) of blastocysts, 25 (76%) diploid- aneuploid mosaic and 3 (9%) aneuploid mosaic, while just 3(9%) of embryos were diploid and 2 (6%) were triploid. Frequency of embryos with more than 50% diploid cells in day-6 were higher than day-7 reanalyzed embryos (8 vs. 3, p<0.05) and hatching/hatched blastocysts didn’t show better self-correction than earlier stages.
Conclusion: While mosaicism is highly frequent in blastocysts, longer culture not only doesn’t lead to better self-correction of aneuploid ies but also might increase the rate of aneuploid cells.
