EMBRYOLOGY: EVALUATION OF IN VITRO FERTILIZATION RATE, EMBRYO QUALITY AND DEVELOPMENT IN EXPERIMENTAL MODEL OF DIABETES INDUCED BY STZ: IN FEMALE MOUSE

Abstract: 

Background: Diabetes mellitus is a metabolic condition characterized by elevated blood glucose levels secondary to absolute impairment of insulin secretion. Maternal diabetes has been demonstrated to adversely affect pre implantation embryo development and pregnancy outcomes. Emerging evidence has implicated that these effects are associated with compromised oocyte competence. Several developmental defects during oocyte maturation in diabetes have been reported over past decades. The objective of the present study was to verify if there is any relation between diabetes mellitus and female fertility. In this study fertility index, in vitro fertilization, and embryo developmental rate were measured.
Materials and Methods: In this study Female mice were injected intraperitoneally with multiple low doses of STZ at 50 mg/kg daily for 5 days to induce type 1 diabetes and diabetes was confirmed 5-7 days after the last injection of STZ by the determination of glycemia. Control sham group received solvent (baffer phosphate 0.1 M) of the drug at same amount and direction. following 30 days of the last injection to induce superovulation in mouse of each group undergoing for gonadotrophin treatment(PMSG and hCG).The oocyte collected from each superovulated mice were fertilize by fresh mouse sperm in HTF medium containing 4mg/ml BSA. The rate of fertilization, two cell embryos, arrested embryos, blastocysts and embryos quality was examined in period of 120 hours. The data were later compared and statistically analyzed by 2Proportion test (p<0.05).
Results: The results obtained from this study revealed that the number of oocyte collected from each mouse in diabetic groups in compared with the control group have significantly decreased and had lower embryo quality. After in vitro fertilization, the fertilization rate, preimplantation embryo development, embryo quality, percentage of two cell embryos and blastosystes in experimental diabetic group have been decreased significantly in compared with control group whereas embryo arrest rate and types of arrested embryo has been increased in experimental group (p<0.05).
Conclusion: Diabetic patient undergoing ovarian stimulation for in vitro fertilization (IVF) are at higher risks of impaired oocyte developmental competence, implantation failure and pregnancy loss. The present study showed that experimental diabetes decreased in oocyte quality, fertilization rate, embryo development and embryo quality.
