DETECTION OF HISTAMINE LEVEL AND RELATION WITH MICROBIAL LOAD OF WHOLE TIGER CROAKER (OTOLITHES RUBER) DURING ICE STORAGE

Abstract: 

In this study, the biogenic amine Histamine (BA) content of whole croaker (Otolithes ruber) was monitored during ice storage for a period of 18 days and related bacterial changes (psychrophiles and mesophiles) were studied every 3 days (0, 3, 6, 9, 12, 15 and 18th day). Histamine was not detected in the first storage day by HPLC and this amine detected after 5 days. The best correlation (R2=0.98) were found among Histamine and mesophilic bacterial load also showed significant variation (P<0.05) during ice storage. Initial concentration of Histamine was 1.49 mg/g and finally reached to 14.21 mg/g when mesophilic bacterial load reached over 107cfu/g. The level of Histamine seemed to be a suitable indicator of freshness of Otolithes rubber.
