SUBEGNOMIC RNAS OF JOIINSON GRASS CHLOROTIC STRIPE MOSAIC VIRUS

Abstract: 

Johnson grass chlorotic stripe mosaic virus (JCSMV) is a small polyhedral virus with a single-stranded positive sense RNA genome of abou t 4.4kb. Dou ble stranded RN A preparations from maize plants infected with JCSMV contained three species with estimated sizes of 4.4, 1.8 and 0.9 kbp. cDNA of 5´ and 3´ ends of JCSMV genome were synthesized, radio labeled and used as probes. Northern blot hybridization analyses of total RNA from infected plants, total RNA from non-infected plants, viral RNA and dsRNAs, showed that genomic RNA reacted with both probes whereas the other two species (1.8kb and 0.9kb) reacted only with 3´ -end probe and are considered to be sub genomic RNA,>.Based on expected number of amino acids encoded by ORP 3 and the size of CP, it is suggested that the large sub genomic RNA (sg RNA1) codes for the coat protein. Sub genomic RNA2 (0.9kb) encodes proteins of ORP4 and ORP5.
