Abstract: 

Isolates of Wheat streak mosaic virus were collected from various parts of Iran and identified serologically. The nucleotide sequence of 3' -untranslated region (3'-UTR) of the genome of each virus isolate was determined as follows: Viral RNA was trapped using a mRNA Capture Kit and complementary DNA was obtained with pligo dT primer (RCFI). A 251 bp DNA segment was amplified by PCR using the cDNA and specific primer pair, WSMI/WSM2. The PCR products were inserted into pTZ57R/f plasmid and cloned in E. coli DH5a. The plasmids were extracted and the inserts sequenced and assessed by Blast search. A neighbour-joining majority-rule consensus phylogram was generated by CLUSTAL X and nucleotide similarity was calculated by Meg Align program.
Depicted phylogram showed that all WSMV isolates could be grouped into four clades. In this analysis, the Iranian isolates were separated into two distinct groups. Isolates from central and southern plateau of Iran formed a distinct clade and the isolates from northwestern Iran categorized with the European isolates in another clade. On the basis of phylogenetic analysis, there are two distinct lineages in Iran, the northwestern lineage and the lineage of central and southern Iranian plateau. The data show that all WSMV strains originated from a common ancestor, probably in Fertile Crescent and diverged into evolutionary lines by geographical isolation. This finding can help find resistance genes for breeding programs. The origin and spreading of WSMV has been discussed.
