THE EXPRESSION OF GUS (GLUEURONIDASE) GENE IN PETUNIA

Abstract: 

Plant breeding is usually conducted via pollination. This method is time-consuming and sometimes impossible. Considering human's need to plant products, one of the suitable methods for gene transformation to plant is using Agrobacterium. In this method, genes can be transferred into many dicious plants. In the present study, Ti–plasmid containing GUS gene linked to NPT II gene under 355 promoter as a construct was supplied. Then transformation in petunia under in vitro condition was carried out using Ti-plasmid. The optimum condition for plant regeneration using MS medium containing 0.5 mg/lit BAP was identified. Then plasmid-prep for presence pZM1047 in Agrobacterium cells was applied. Finally, co-culture of Agrobacterium with petunia explants for gene transformation was carried out and transgenic plants with 5% frequency were obtained. PCR amplification confirmed presence of GUS gene in transgenic plants.
